Aktywność dysmutaz ponadtlenkowych i katalazy oraz fluorescencja chlorofilu in vivo w odpowiedzi na zmianę natężenia światła w czasie rozwoju siewek A. thaliana

Działanie stresu świetlnego na rośliny jest coraz lepiej poznane, jednakże dalej brakuje danych na temat reakcji na ten czynnik stresowy u roślin w stadium siewki. Niewiele też wiadomo na temat wpływu stresu świetlnego na siewki poddane wstępnie etiolacji i następnie deetiolowane. Jako model badawczy w niniejszej pracy wykorzystano 7- dniowe siewki A.thaliana hodowane w ciągłym świetle białym (określane w pracy jako „zielone”) oraz poddane 3-dniowej etiolacji i następnie deetiolowane przez 4 dni (określane w pracy jako „deetiolowane”). Głównym celem pracy było zbadanie wpływu stresu świetlnego na funkcjonowanie aparatu fotosyntetycznego oraz na aktywność dysmutaz ponadtlenkowych (SOD) oraz katalazy (CAT). Rośliny poddawano krótkotrwałej (45 lub 90 min) ekspozycji na światło białe o wysokim natężeniu (850, 1300, 2000 µmol·m-2·s-1), a następnie inkubowano je przez określony czas (0, 2, 4, 24 h) w warunkach hodowli. Pomiary aktywności katalazy wykonywano metodą spektrofotometryczną. Aktywność SOD oznaczano w żelu elektroforetycznym wykorzystując reakcję hamowania redukcji NBT na skutek dysmutacji anionorodnika ponadtlenkowego przez SOD. Wpływ stresu świetlnego na działanie aparatu fotosyntetycznego zmierzono wykorzystując technikę obrazowania fluorescencji chlorofilu in vivo. Przed wykonaniem badań głównych przeprowadzono szereg optymalizacji: długości czasu deetiolacji siewek oraz ilości białka do przeprowadzenia pomiarów aktywności enzymatycznej CAT oraz SOD. Wykazano, że badane warunki stresowe mają niewielki wpływ na aktywność aparatu fotosyntetycznego oraz na aktywność enzymatyczną SOD oraz CAT. Na podstawie analizy krzywych zmian NPQ zaproponowano występowanie kilku populacji wśród siewek deetiolowanych, różniących się między sobą czasem osiągnięcia maksimum wygaszania niefotochemicznego, niezależnie od warunków środowiska. Przeprowadzone badania mają charakter wstępny i należałoby je powtórzyć z uwzględnieniem większej liczby powtórzeń biologicznych.
The effect of light stress on plants is better and better known. However, data on the response to this stress factor in plants at the seedling stage are still incomplete. Little is known about the effect of light stress on pre-etiolated and then deetioled seedlings. In this study, 7-day-old A. thaliana seedlings were used as a research model. They were either grown in continuous white light (referred in the study as „green”) or subjected to a 3-day etiolation and then deetiolated for 4 days (referred in the study as „deetiolated”) The main aim of the study was to investigate the effect of light stress on the function of the photosynthetic apparatus, and on the activity of superoxide dismutases (SOD) and catalase (CAT). Plants were exposed to a short-term (45 or 90 min) illumination with white light of various high intensities (850, 1300, 2000 µmol m-2 s-1), and then they were incubated for a specified time (0, 2, 4, 24 h) under the culture conditions. Catalase activity was measured using the spectrophotometric method. SOD activity was determined in electrophoretic gels using the inhibition of the reaction of NBT reduction due to superoxide radical dismutation by SOD. In vivo chlorophyll fluorescence imaging technique was used to assess the effect of the light stress on the function of the photosynthetic apparatus Before the main experiments were performed, a number of optimizations were carried out: the length of the seedling deetiolation time, and the amount of protein needed to measure the enzymatic activity of CAT and SOD. It was shown that the tested stress conditions had little effect on the activity of the photosynthetic apparatus and on the enzymatic activity of SOD and CAT. Based on the analysis of NPQ curves, it was proposed that there were several populations among deetiolated seedlings, differing in the time needed to reach the maximum of the non-photochemical extinction, regardless of environmental conditions. The conducted research is preliminary and should be repeated with a larger number of biological repetitions.
