Results transcriptomes sequencing dataset for human melanoma and melanocytes cell cultures

Results of totalRNA transcriptome sequencing for human melanoma: WM115, WM266-4, and melanocytes HEMa-LP cell lines in (*.xlsx) files format. The raw readings were mapped to the human reference genome GRCh38 from the Ensembl database (v. 96) using the Hisat2 v. 2.1.0 program (2). Cuffquant and Cuffmerge version 2.2.1 (3) and the GTF Homo_sapiens.GRCh38.96.gtf file from the Ensembl database were used to calculate the expression level of the transcripts. In the presented analysis, 131 621 transcripts were measured. A one-way ANOVA analysis was performed aimed at identifying transcripts with significantly different expression levels in at least one group experimental. The obtained p-values were corrected for multiple testing using the FDR algorithm For transcripts with significantly different expression levels in the studied groups (FDR from test ANOVA <0.1) post hoc analysis was performed with Student's t-test. Each group was compared with each, and the obtained p-values were corrected by the Bonferroni correction.
Celem niniejszej pracy było utworzenie zbioru transkryptomicznych danych dla linii czerniaka skóry o zróżnicowanym stopniu złośliwości oraz melanocytów. Wykorzystano trzy modele linii komórkowych: WM115 pierwotna linia w fazie wzrostu pionowego; WM266-4 przerzut z węzłów chłonnych w fazie wzrostu pionowego, jako przykład linii nowotworowych oraz linię melanocytów – prawidłowych komórek skóry. Banki komórkowe WM115, WM266-4, pochodzą z the European Searchable Tumour Cell Line and Data Bank (ESTDAB)- A Collection of Immunologically Characterised Melanoma Cell Lines and Databank (Tübingen, Germany). Bank linii melanocytów HEMa-LP zostały uzyskane przez Life Technologies, Paisley, UK. Dane zostały wygenerowanie z użyciem platformy sekwencjonowania nowej generacji Illumina.
The aim of this study was to create a set of transcriptomics data for skin melanoma lines of various degrees of malignancy and for normal skin cells - melanocytes.
