abstract in Polish:	SNAI1 to ludzkie białko pełniące rolę represora transkrypcji, należące do rodziny czynników transkrypcyjnych zawierających palce cynkowe. SNAI1 pełni istotne funkcje w rozwoju zarodkowym, regulując takie procesy jak formowanie się mezodermy czy grzebienia nerwowego. Rola SNAI1 w dorosłym organizmie badana była do tej pory głównie w kontekście funkcji, jaką pełni to białko w procesie przerzutowania komórek nowotworowych poprzez regulację przejścia epitelialno-mezenchymalnego. Najnowsze doniesienia sugerują również wpływ Snai1 na różnicowanie mięśniowe mysich mioblastów. Głównym celem niniejszej pracy było stworzenie dogodnego modelu miogenezy in vitro do dalszego badania roli SNAI1 w różnicowaniu mięśniowym ludzkich komórek.W tym celu w pierwszym etapie badań zoptymalizowano dwa literaturowe protokoły różnicowania ludzkich indukowanych pluripotencjalnych komórek macierzystych (komórek iPS) do komórek mięśni szkieletowych. Obydwie testowane metody umożliwiły uzyskanie komórek wykazujących morfologię mięśni szkieletowych oraz ekspresję markerów molekularnych charakterystycznych dla tych komórek. W trakcie procesu różnicowania zaobserwowano także znaczący spadek ekspresji SNAI1, co sugeruje udział tego czynnika transkrypcyjnego w regulacji miogenezy. Aby zbadać rolę SNAI1 w różnicowaniu mięśniowym komórek iPS zdecydowano się na wyciszenie genu SNAI1 w tych komórkach za pomocą nukleaz CRISPR/Cas9 i TALEN. W tym celu przetestowano efektywność działania tych dwóch typów nukleaz na wykazujących ekspresję SNAI1 komórkach mięsaka prążkowanokomórkowego. Pozwoliło to na wybór najbardziej efektywnej metody do modyfikacji poziomu SNAI1 w komórkach iPS w przyszłości. Równocześnie zdecydowano się na otrzymanie komórek iPS z nadekspresją SNAI1. W ostatnim etapie badań przetestowano wpływ SNAI1 na różnicowanie ludzkich mioblastów. Podsumowując, wyniki uzyskane w pracy umożliwiają badanie roli SNAI1 w regulacji miogenezy dzięki otrzymaniu modelu odwzorowującego ten proces in vitro oraz dzięki optymalizacji wykorzystania metod molekularnych służących do trwałej modyfikacji ekspresji SNAI1 w komórkach iPS, a także wskazują na ważną rolę SNAI1 w regulacji różnicowania ludzkich mioblastów.
abstract in English:	SNAI1 is a transcription repressor belonging to family of zinc-finger transcription factors. SNAI1 plays a crucial role in embryogenesis by regulating processes such as formation of mesoderm and development of neural crest. The role of SNAI1 in an adult organism was investigated mainly in an induction of metastasis through epithelial-mesenchymal transition during tumor development. Recent data demonstrated also the important role of Snai1 in myogenic differentiation of murine myoblasts. The major objective of this study was to establish a model of iPS-derived myogenesis in order to investigate the effect of SNAI1 on human myogenesis. To accomplish that goal the appropriate model of myogenesis in vitro had to be established. Therefore, two literature protocols of myogenic differentiation of human induced pluripotent stem (iPS) cells were optimized. Both of the tested methods led to generation of cells displaying morphology of skeletal muscle cells and expression of molecular markers of those cells. Moreover, during myogenic differentiation the level of SNAI1 was decreasing, what suggests that it may play a significant role in this process. To further investigate the role of SNAI1 in myogenesis it was decided to either downregulate or upregulate its expression level with molecular methods. Therefore, two sets of experiments have been performed. Firstly, the genome editing technology using CRISPR/Cas9 and TALEN nucleases has been established. Rhabdomyosarcoma cell line, expressing SNAI1 was used as a model to test and select the most efficient nucleases for further studies on genome editing in iPS cells. Secondly, iPS cell line displaying stable overexpression of SNAI1 was established. In parallel, the effect of SNAI1 on myogenic differentiation of human myoblasts was investigated. To conclude, in preparation of this thesis the methodology to study the human myogenesis using iPS cells as a starting point was established. Moreover, the molecular technologies enabling modification of SNAI1 expression in iPS cells were established and optimized. Additionally, the experimental results that have been obtained suggest that SNAI1 might play a crucial role in regulation of human myogenesis.In the future these results warrant further investigation of this phenomenon using the established iPS myogenic differentiation model in vitro and the optimized molecular methods for stable modifications of SNAI1 expression level in iPS cells.
