Poszukiwanie zależności pomiędzy strukturą a aktywnością biologiczną ligandów wybranych receptorów serotoninowych

W niniejszej pracy przedstawiono próbę identyfikacji cech strukturalnych ligandów z klasy długołańcuchowych arylopiperazyn (LCAP), istotnych ze względu na wiązalność do ludzkich receptorów serotoninowych: 5-HT 1A, 5-HT 2A i 5-HT 7. Badania oparto na analizie 11 struktur krystalicznych LCAP. Wybrane pochodne zadokowano do modeli homologicznych wymienionych receptorów. Parametry geometryczne fragmentu fenylopiperazyny porównano z tymi, obecnymi w strukturach zdeponowanych w Cambridge Structural Database. Otrzymane wyniki zestawiono z danymi powinowactwa badanych związków do receptorów 5-HT 1A, 5-HT 2A i 5-HT 7, podejmując próbę wyjaśnienia obserwowanych różnic w ich aktywności. Opracowano również protokół, pozwalający na przeprowadzenie wirtualnych badań przesiewowych milionów związków chemicznych na pojedynczej stacji roboczej w przeciągu kilku dni. Protokół oparto na dokowaniu molekularnym oraz porównywaniu kształtu ligandów. Protokołu użyto do poszukiwań ligandów receptora 5-HT 7, nie wykazujących powinowactwa do receptora 5-HT 1A. Część najwyżej ocenionych związków zakupiono. W oparciu o uzyskane dlannich wyniki stałych powinowactwa, przeprowadzono dyskusję efektywności zaproponowanego protokołu.
This thesis presents an attempt to identify the structural features of long-chain arylpiperazine (LCAP) ligands needed for achieving high affinity towards human 5-HT 1A, 5-HT 2A i 5-HT 7 serotonin receptors. The study was based on the analysis of 11 LCAP crystal structures obtained in this work. Selected derivatives were also docked to homology models of the said receptors. The geometrical parameters of the phenylpiperazine fragments were compared with those in the structures deposited in the Cambridge Structural Database. The obtained results were confronted with the differences observed in the binding affinities towards 5-HT 1A, 5-HT 2A i 5-HT 7 receptors in an attempt of explaining them. The work also describes a novel protocol that allows to perform a virtual screening of millions of compounds on a single workstation within a few days. The protocol was based on two methods: molecular docking and ligand shape comparison. The protocol was used to find 5-HT 7 ligands that would not bind to the 5-HT 1A receptor. Some of the top scored compounds were purchased and their inhibition constants were determined. The effectiveness of the proposed protocol was discussed based on the obtained results.
