Badanie fotoprzełączalnej cytotoksyczności i oddziaływania z biomolekułami przeciwnowotworowego kompleksu platyny(II)

dc.abstract.enThe aim of the study was to determine the difference in cytotoxicity between the photoswitchable anticancer cisplatin analog cis-PtCl2(PS1)2, as a result of intracellular photoisomerization. Studies were performed on cancer cell lines of human melanoma (B16-F10), human prostate (PC3) and mouse mammary gland cancer (4T1) and also on a normal mouse mammary gland cell line (NMuMG). The study showed the possibility of photoswitching of the complex after internalization by the cells, which resulted in an increase in their survival rate due to trans-cis photoisomerization. Similarly, cis-trans photoisomerization of the internalized complex under visible light (530 nm) resulted in increased cytotoxicity of the complex, which led to decreased cell survival. In the case of the normal NMuMG cell line, the cis-PtCl2(cis-PS1)2 photoisomer caused no apparent cytotoxic effect, while the trans photoisomer significantly reduced cell survival. The trans-cis photoisomerization of the internalized complex after 30 min of incubation of the compound with cells resulted in a change in the cytotoxicity of the complex, which is of great importance in the context of using the tested compound as a potential therapeutic agent. A next step of the study verified whether isomers of the photoswitchable analog of cisplatin show similar affinity for N7 guanine as cisplatin, one of whose main mechanisms of action is its covalent binding to DNA nucleotide bases, preferentially to the N7 guanine atom. [2] Previous studies have shown that the complex being studied interacts with DNA. [1] However, studies using 1H NMR spectroscopy were unable to confirm such an interaction, although they suggest a different kind of interaction. The study also assessed the ability of both photoisomers of the complex to generate reactive oxygen species, which is an important mechanism of the cytotoxic action of classical cisplatin. [2] The study confirmed the ability of the studied complex to generate hydroxyl radicals and singlet oxygen, which can have a significant role in the induction of oxidative stress in cells.pl

Badania przeprowadzono na nowotworowych liniach komórkowych ludzkiego czerniaka (B16-F10), ludzkiej prostaty (PC3) oraz mysiego nowotworu gruczołu sutkowego (4T1) a także na prawidłowej linii komórkowej mysiego gruczołu sutkowego (NMuMG). Badania wykazały możliwość fotoprzełączania kompleksu po internalizacji przez komórki, które skutkowało wzrostem ich przeżywalności w wyniku fotoizomeryzacji trans-cis. Podobnie fotoizomeryzacja cis-trans zinternalizowanego kompleksu pod wpływem światła widzialnego (530 nm) powodowała zwiększenie cytotoksyczności kompleksu, co w konsekwencji prowadziło do zmniejszenia przeżywalności komórek. W przypadku prawidłowej linii komórkowej NMuMG fotoizomer cis-PtCl2(cis PS1)2 nie spowodował widocznego efektu cytotoksycznego, natomiast fotoizomer trans wyraźnie obniżał przeżywalność komórek. Fotoizomeryzacja trans cis zinternalizowanego kompleksu po 30 min inkubacji związku z komórkami powodowała zmianę cytotoksyczności kompleksu, co ma ogromne znaczenie w kontekście zastosowania badanego związku jako potencjalnego środka terapeutycznego. W kolejnym etapie badań zweryfikowano czy izomery fotoprzełączalnego analogu cisplatyny wykazują podobne powinowactwo do N7 guaniny jak cisplatyna, której jednym z głównych mechanizmów działania jest jej kowalencyjne wiązanie z zasadami azotowymi DNA, preferencyjnie z atomem N7 guaniny. [1] Wcześniejsze badania wykazały, że badany kompleks oddziałuje z DNA. [2] Badania za pomocą spektroskopii 1H NMR nie pozwoliły jednak potwierdzić takiej interakcji, choć wskazują na innego rodzaju oddziaływanie. W ramach pracy przeprowadzono również ocenę zdolności obu fotoizomerów badanego kompleksu do generowania reaktywnych form tlenu, które stanowi istotny mechanizm cytotoksycznego działania klasycznej cisplatyny. [1] Badania pozwoliły potwierdzić możliwość generowania rodników hydroksylowych oraz tlenu singletowego przez badany kompleks, które mogą odgrywać istotną rolę w indukowaniu stresu oksydacyjnego w komórkach.pl
