dc.abstract.enSpexin (SPX) is a neuropeptide discovered in 2007. It is acting through the GALR2 and GALR3 receptors, which expression has been described in central and peripheral tissues of many vertebrate species. Its pleiotropic effect on the body has also been demonstrated. SPX is taking part in body weight regulation, affects lipid storage and inflammation. Moreover, it is taking part in many pathological conditions, including obesity, diabetes, gestational diabetes and pre-eclampsia. The development of the human placenta is a very dynamic and complicated process, influenced by many factors. Studies have shown that adipokines, such as leptin, apelin and adiponectin are produced by placental cells and affect their proliferation and endocrine functions. To date, there is no data on the effect of SPX in the functioning of human placental cells. Therefore, this bachelor's thesis was aimed to study the expression of SPX and GALR2 and GALR3 receptors in human placental cells and to determine the effect of SPX on placental cell proliferation. The research model used in the experiments was based on two trophoblast lines - the JEG-3, line corresponding to the cells of the extra-villous cytotrophoblast and the BeWo line, corresponding to the cells of the villous cytotrophoblast, which has the ability to differentiate into syncytiotrophoblast. The cells were placed in the appropriate medium and after 24, 48 and 72 h of culture, the test material was collected to examine the expression of the SPX. GALR2 and GALR3 genes were tested using real time PCR method, and the protein level was determined by Western Blot. To determine the effect of SPX on cell proliferation, SPX was added to the JEG-3 and BeWo cultures at doses of 0.1, 1, 10 and 100 nM, followed by the alamarBlue assay after 24, 48 and 72 h. The results were analyzed statistically in GraphPad Prism 8.0.1 using the one-way ANOVA test. The conducted studies showed the expression of SPX as well as GALR2 and GALR3 in human placental cells. Moreover, in the JEG-3 line after 24 and 48 h, and in the BeWo line, the inhibitory effect of SPX on the proliferation of trophoblast cells was demonstrated at all times tested. The obtained study results indicate for the first time the expression of mRNA and protein for SPX and GALR2 and GALR3 receptors in human placenta cells and the participation of SPX in the proliferation of trophoblast cells. Further studies are necessary to confirm the involvement of SPX in the regulation of placental cell function and to confirm its involvement in the development of the human placenta.pl

Wykazano także jej plejotropowy wpływ na organizm. SPX bierze udział w regulacji masy ciała, wpływa na magazynowanie lipidów oraz stany zapalne. Co więcej, bierze udział w wielu stanach patologicznych, do których należy otyłość, cukrzyca, cukrzyca ciążowa, czy stan przedrzucawkowy. Rozwój łożyska ludzkiego jest bardzo dynamicznym i skomplikowanym procesem, na który wpływ ma wiele czynników. Badania wykazały, że adipokiny takie jak leptyna, apelina czy adiponektyna produkowane są przez komórki łożyska oraz wpływają na ich proliferację i funkcje endokrynne. Dotychczas brak jest danych dotyczących wpływu SPX na funkcjonowanie komórek łożyska ludzkiego. Dlatego też celem niniejszej pracy licencjackiej było zbadanie ekspresji SPX oraz receptorów GALR2 i GALR3 w komórkach łożyska ludzkiego oraz określenie wpływu SPX na proliferację komórek łożyska. Modelem badawczym wykorzystanym w eksperymentach były dwie linie trofoblastu - linia JEG-3 odpowiadająca komórkom cytotrofoblastu pozakosmkowego oraz linia BeWo odpowiadająca komórkom cytotrofoblastu kosmkowego, który ma możliwość różnicowania się do syncytiotrofoblastu. Komórki wysiewano w odpowiednim medium i po 24, 48 i 72 h hodowli materiał do badań zebrano w celu określenia ekspresji genu SPX, GALR2 i GALR3 za pomocą metody real time PCR, natomiast poziom białka oznaczono za pomocą Western Blot. W celu określenia wpływu SPX na proliferację komórek, do hodowli JEG-3 i BeWo dodano SPX w dawkach 0,1, 1, 10 i 100 nM, a następnie po 24, 48 i 72 h przeprowadzono test alamarBlue. Otrzymane wyniki poddano analizie statystycznej w programie GraphPad Prism 8.0.1 z wykorzystaniem testu jednoczynnikowej ANOVA. Przeprowadzone badania wykazały ekspresję SPX oraz GALR2 i GALR3 w komórkach łożyska ludzkiego. Ponadto, w linii JEG-3 po czasie 24 i 48 h, a w linii BeWo we wszystkich badanych czasach wykazano hamujące działanie SPX na proliferację komórek trofoblastu. Uzyskane badania po raz pierwszy wskazują na ekspresję mRNA i białka dla SPX i receptorów GALR2 i GALR3 w komórkach łożyska ludzkiego oraz udział SPX w proliferacji komórek trofoblastu. Dalsze badania potwierdzające udział SPX w regulacji funkcji komórek łożyska są niezbędne aby potwierdzić jej udział w rozwoju łożyska ludzkiego.pl
