Analiza zmienności i struktury genetycznej populacji Potamogeton rutilus – gatunku rzadkiego w Europie i zagrożonego wyginięciem w Polsce

dc.abstract.enPotamogeton rutilus is a Central European with one of the smallest area of occurrence among the representatives of the genus. In Poland, it is known from the lake Rotcze at Lubelszczyzna and the lakes: Białe, Rospuda i Kamienne at Suwalszczyzna. During the last decades Potamogeton rutilus became extinct at several localities in the western part of the range (Germany, Poland). The aim of the present study was an analysis of the genetic variation of P. rutilus and its spatial structure. The study was conducted on 67 individuals from eight populations in Poland, Lithuania and Estonia. The analysis of the AFLP profiles, generated in automatic sequencer, were scored using Genographer 2.1. The analysis of polymorphism (namely number of polymorphic markers, the Nei coefficient of genetic variation and clonal variability of the population) was performed using the AFLPdat. Variation within and among populations and its spatial structure was estimated based on: (i) the scatterdiagrams PcoA analysis (program MVSP), (ii) Net Neighbor Network (SplitsTreeprogram), (iii) Bayesian analysis of genetic structure (the program STRUCTURE), (iv) analysis of molecular variance – AMOVA (Arlequin program). The observed genetic structure indicates that a high genetic isolation of individual populations exists, presumably associated with low efficiency of sexual reproduction and recruitment of new individuals as a result of generative processes, and thus, little genetic variability of populations and gene flow among small populations occurs. Results of the study can be used for optimal conservatory activities protecting the global resources of Potamogeton rutilus.pl

Rotcze na Lubelszczyźnie oraz jezior: Białe, Rospuda, i Kamienne na Suwalszczyźnie. Jak wynika z monitoringu gatunku, w ostatnich dziesięcioleciach takson ten wymarł na kilku stanowiskach w zachodniej części zasięgu (Niemcy, Polska).Celem pracy było zbadanie zmienności genetycznej Potamogeton rutilus oraz jej przestrzennej struktury aby móc efektywniej chronić zasoby tego gatunku.Badania przeprowadzono na 67 osobnikach z ośmiu populacji z Polski, Litwy i Estonii. Do analizy wykorzystano technikę AFLP a otrzymane profile, wygenerowane na sekwencjonerze automatycznym, analizowano za pomocą programu Genographer 2.1. Obliczono poziom polimorfizmu markerów wyrażony poprzez: liczbę markerów polimorficznych, współczynnik zmienności genetycznej Nei’a, zmienność klonalną populacji, z zastosowaniem programu AFLPdat. Zmienność wewnątrz- i międzypopulacyjną oraz jej przestrzenną strukturę analizowano za pomocą: (i) diagramów rozrzutu analiz PcoA (program MVSP); (ii) sieci Neighbor Net (program SplitsTree); (iii) bayerowskich analiz struktury genetycznej (program STRUCTURE); (iv) analizy wariancji molekularnej – AMOVA, (program Arlequin). Zaobserwowana struktura genetyczna świadczy o dużej izolacji genetycznej poszczególnych populacji, związanej przypuszczalnie z niewielką efektywnością rozmnażania generatywnego, i co z tym związane, znikomą rekrutacją młodych osobników, powstałych wyniku procesów płciowych. W populacjach realizowane jest głównie rozmnażanie wegetatywne czego wynikiem jest niewielka zmienność genetyczna populacji i niewielki przepływ genów między populacjami.pl
