dc.abstract.enThe aim of this study was to investigate the effect of multiple administrations of zinc oxide nanoparticles and zinc oxide nanoparticles together with ketoprofen for the selected parameters of oxidative stress: superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) in vivo. The impact of nanoparticles on the changes caused by administration of ketoprofen was also determined. The studies were carried out on the serum, liver and kidney samples taken from rats.The studies have shown that there is no substantial peroxidation of lipids in the liver and kidney on exposure on zinc oxide nanoparticles. Thus it was concluded that the nanoparticles do not damage the cell membranes, so one can consider to use them to enhance mobilization of antioxidant barrier.Nanoparticles of zinc oxide do not induce of the oxidative stress in the liver and kidney, as evidenced by an increase in GSH concentration in these organs. In contrast, there was no statistically significant changes in the concentration of glutathione in serum in comparison with the control group. Nanoparticles probably affect the stimulation of the activity of antioxidant barrier by increasing the activity of SOD in the liver and serum. On the other hand, the kidney showed no statistically significant differences in SOD activity as compared with the control group. When ketoprofen was applied there was no intensification of the process of lipid peroxidation in liver and kidney.Based on the results obtained it can be clearly stated that the use of nanoparticles of zinc oxide increases the activity of antioxidant barrier by increasing the production of SOD and GSH. ZnO nanpoarticles do not disrupt cellular homeostasis because they do not disrupt the cell membranes.pl
dc.abstract.plPrzedmiotem niniejszej pracy było zbadanie wpływu wielokrotnych podań nanocząstek tlenku cynku i nanocząstek tlenku cynku z ketoprofenem na wybrane parametry stresu oksydacyjnego: dysmutazę ponadtlenkową (SOD), glutation zredukowany (GSH) oraz dialdehyd malonowy (MDA) w badaniach in vivo. Określono także wpływ nanocząstek na zmiany wywołane podaniem ketoprofenu. Materiał badawczy stanowiła surowica, wątroba i nerka pobrane od szczurów.Przeprowadzone badania z zastosowaniem nanocząstek tlenku cynku nie wykazały nasilenia procesów peroksydacji lipidów w wątrobie i nerce. Na tej podstawie można wnioskować, że nanocząstki nie powodują uszkodzenia błon komórkowych, więc można je z powodzeniem stosować w celu zwiększenia mobilizacji bariery antyoksydacyjnej.Nanocząstki tlenku cynku nie przyczyniały się do rozwoju stresu oksydacyjnego w wątrobie i nerce, o czym świadczy wzrost stężenia GSH w tych narządach. Natomiast w surowicy nie wykazano istotnych statystycznie zmian w stężeniu glutationu w porównaniu z grupą kontrolną. Nanocząstki prawdopodobnie wpływały na stymulację aktywności bariery antyoksydacyjnej, poprzez podnoszenie aktywności SOD w wątrobie i w surowicy. Z kolei w nerce nie wykazano istotnych statystycznie różnic w aktywności SOD w porównaniu z grupą kontrolną. Podczas stosowania ketoprofenu nie wykazano nasilania procesu peroksydacji lipidów w wątrobie i nerce. Na podstawie uzyskanych wyników badań można stwierdzić, że stosowanie nanocząstek tlenku cynku podnosi aktywność bariery antyoksydacyjnej, poprzez zwiększenie produkcji SOD oraz GSH. Nanocząstki ZnO nie zaburzają homeostazy komórkowej, gdyż nie wykazują działania niszczącego błony komórkowe.pl
