Teoretyczne badania wpływu czerwieni Kongo na pasywny transport doksorubicyny przez modelowe błony komórkowe
Interactions of doxorubicin-Congo red complexes with model cell membranes: a molecular dynamics study

Doksorubicyna jest jednym z najczęściej stosowanych leków przeciwnowotworowych, a jej zastosowanie kliniczne ogranicza silna kardiotoksyczność. Czerwień Kongo, barwnik diazowy zdolny do samoorganizacji w roztworach wodnych, tworzy z doksorubicyną supramolekularny kompleks, co czyni ją potencjalnym kandydatem na nośnik leku. Celem pracy była charakterystyka kompleksów czerwieni Kongo z doksorubicyną oraz ich oddziaływań z modelowymi błonami biologicznymi, wykorzystując metody klasycznej dynamiki molekularnej. Zbadano cztery formy protonacyjne doksorubicyny współistniejące w pH fizjologicznym, samą czerwień Kongo oraz układy mieszane, w dwóch modelach dwuwarstwy lipidowej odwzorowujących skład błony komórki prawidłowej i nowotworowej. Choć oba modele różnią się przede wszystkim stopniem uporządkowania i sztywnością, o powinowactwie badanych cząsteczek do powierzchni decyduje ładunek błony. Formy leku zawierające protonowaną grupę aminową adsorbują się na anionowej błonie nowotworowej, natomiast podwójnie ujemnie naładowana czerwień Kongo jest od tej powierzchni odpychana i wiąże się wyłącznie z obojętną elektrycznie błoną prawidłową. Wszystkie cztery formy leku tworzą z barwnikiem trwały kompleks, powstający samorzutnie w fazie wodnej, utrzymywany głównie przez oddziaływanie pomiędzy płaskimi układami aromatycznymi, uzupełnione kontaktem elektrostatycznym między grupą aminową a sulfonową. Porównanie obu form zwitterjonowych wykazało, że o zachowaniu cząsteczki decyduje nie ładunek wypadkowy, lecz jego rozmieszczenie.Wpływ kompleksowania na powinowactwo leku do błony okazał się przeciwny w obu modelach. Na błonie prawidłowej kompleks adsorbuje się w połowie przypadków, podczas gdy sama doksorubicyna prawie wcale. Na błonie nowotworowej obecność barwnika niemal całkowicie znosi powinowactwo leku, a jego adsorpcja wymaga uprzedniej dysocjacji kompleksu. Barwnik narzuca zatem całemu kompleksowi własną preferencję. Dodatkowo kompleks jest zakotwiczony w błonie przez barwnik, a nie przez lek. Uzyskane wyniki wskazują, że kompleksowanie z czerwienią Kongo odsuwa doksorubicynę od anionowej powierzchni błony, co jest rezultatem przeciwnym do oczekiwanego na podstawie badań eksperymentalnych.
Doxorubicin is among the most widely used anticancer drugs, yet its clinical application is limited by severe cardiotoxicity. Congo red, a self-assembling diazo dye, forms a supramolecular complex with doxorubicin and has therefore been proposed as a drug carrier. Experiments have shown that this complex interacts with model lipid membranes more strongly than either component alone, but the underlying molecular mechanism and its dependence on membrane composition have remained unknown. The aim of this study was to characterise Congo red–doxorubicin complexes and their interactions with model biological membranes by means of classical molecular dynamics. Four protonation forms of doxorubicin coexisting at physiological pH, Congo red alone, and mixed systems were investigated in two lipid bilayer models representing the composition of a normal and a cancerous cell membrane. Although the two models differ primarily in their degree of ordering and rigidity, the affinity of the studied molecules for the surface is determined by the charge of the membrane. Drug forms containing a protonated amine group adsorb onto the anionic cancer membrane, whereas the doubly negatively charged Congo red is repelled from this surface and binds exclusively to the electrically neutral normal membrane. All four forms of the drug form a stable complex with the dye, which arises spontaneously in the aqueous phase and is held together mainly by the interaction between the planar aromatic systems, supplemented by an electrostatic contact between the amine and sulfonate groups. A comparison of the two zwitterionic forms showed that the behaviour of the molecule is determined not by its net charge, but by the distribution of that charge.The effect of complexation on the affinity of the drug for the membrane proved to be opposite in the two models. On the normal membrane, the complex adsorbs in half of the cases, while doxorubicin alone adsorbs almost not at all. On the cancer membrane, the presence of the dye almost completely abolishes the affinity of the drug, and its adsorption requires prior dissociation of the complex. The dye therefore imposes its own preference on the complex as a whole. Moreover, the complex is anchored in the membrane by the dye rather than by the drug.The results obtained indicate that complexation with Congo red draws doxorubicin away from the anionic membrane surface, a result opposite to that expected on the basis of experimental studies.
