The dependency of dominance effects on the phylogenetic distance in the natural yeast strains

Większość występujących w przyrodzie mutacji powoduje szkodliwe zmiany, które objawiają się spadkiem dostosowania. Niniejszy eksperyment miał na celu sprawdzenie, w jakim stopniu mutacje szkodliwe są recesywne. W szczególności, czy pozytywny efekt heterozji jest związany z odległością filogenetyczną pomiędzy szczepami Saccharomyces cerevisiae Badano 22 homozygoty oraz 231 diploidów heterozygotycznych, a głównym mierzonym parametrem było maksymalne tempo wzrostu. Wykazano wyższe średnie dostosowanie heterozygot w porównaniu do homozygot. Prawdopodobnie heterozja spowodowana jest przez zwykłą dominację alleli niezmutowanych w jednym genomie, które kompensują szkodliwe efekty w drugim genomie. W związku z tym, że korelacja pomiędzy efektem heterozji, a odległością filogenetyczną jest niska, przypuszcza się, że spadek dostosowania w homozygotach spowodowany jest raczej przez stosunkowo niedawne pojawienie się niewielu dość silnych efektów, niż długotrwałą akumulację wielu małych efektów.
The majority of mutations present in nature have damaging effects, which manifest in a decreased fitness of the organism. The aim of the present research was to check to what extand the harmful mutations are recessive. It was studied in particular, whether the positive effect of heterosis is connected with phylogenetic distance between Saccharomyces cerevisiae strains. We have analysed 22 homozygotes and 231 heterozygous diploids, and the main parameter measured was the maximal growth rate. The research has proved that the average fitness of heterozygotes was higher than of homozygotes. Presumably, heterosis is caused by normal dominance of non-mutated alleles from one genome, which compensate detrimental effects in another genome. Since the correlation between heterosis effect and phylogenetic distance is low, it is assumed that the decrease in fitness in homozygotes is caused by the relatively recent rise of few moderately strong effects rather, than long-term accumulation of many but quite small effects.
