Literatura naukowa na temat biologii mezenchymalnych komórek macierzystych w mięśniówce gładkiej macicy jest dość uboga a obecność komórek pluripotencjalnych w myometrium kontrowersyjna. Ostatnie badania wskazują na obecność odrębnej subpopulacji komórek macierzystych/progenitorowych w myometrium człowieka i myszy, i sugerują ich udział w przebudowie macicy w różnych stanach fizjologicznych. Wielką zaletą świni jako modelu doświadczalnego jest duże podobieństwo anatomiczne i fizjologiczne w organizacji narządów układu rozrodczego do człowieka. Celem niniejszej pracy była próba identyfikacji komórek macierzystych w mięśniówce macicy świni na podstawie ekspresji wybranych znaczników mezenchymalnych komórek macierzystych CD44 i CD146 oraz markerów komórek pluripotencjalnych Oct4, Nanog, Sox2, SSEA4 i TRA-1-60. Uzyskane wyniki nie wykazały obecności takich komórek w mięśniówce macicy świni w fazie proliferacyjnej ani sekrecyjnej cyklu estralnego. Zidentyfikowano natomiast nieliczne, pojedyncze komórki CD44+ oraz CD146+ w strefie łączącej endometrium i myometrium (junctional zone), które stanowić mogą populację domniemanych mezenchymalnych komórek macierzystych. Ponadto stwierdzono wyraźną zależność ekspresji TRA-1-60 oraz SSEA4 w komórkach nabłonkowych endometrium od fazy cyklu płciowego, a tym samym od statusu hormonalnego badanych świń.
The scientific literature on the biology of mesenchymal stem cells in the uterine smooth muscles is quite scarce and the presence of pluripotent cells in the myometrium is controversial. Recent studies indicate the presence of distinct subpopulation of stem/ progenitor cells in the human and mouse myometrium and suggest their involvement in uterine remodeling in various physiological conditions. The great advantage of the pig as an experimental model is the large anatomical and physiological similarity in organization of the reproductive system organs to humans. The aim of this study was to identify stem cells in the porcine uterine muscle on the basis of the expression of selected mesenchymal stem cell markers CD44 and CD146 and markers of pluripotent cells Oct4, Nanog, Sox2, SSEA4 and TRA-1-60. The obtained results did not show the presence of such cells in porcine uterine muscles in the proliferative or secretory phases of the estrus cycle. However, a few single cells CD44+ and CD146+ were identified in the junctional zone, which may constitute a population of putative mesenchymal stem cells. Moreover, there was a clear relationship of TRA-1-60 and SSEA4 expression in endometrial epithelial cells with the phase of the estrous cycle, and thus with the hormonal status of pigs.
