Było to epokowym odkryciem, a także nieprawdopodobnym zaskoczeniem dla środowiska naukowego,gdy powszechnie znane szkodliwe substancje gazowe – tlenek azotu (NO) i siarkowodór (H2S) – okazały się być istotnymi i niezbędnymi molekularnymi elementami ludzkiego metabolizmu i biochemii. W warunkach fizjologicznych pełnią one role wewnątrz- oraz międzykomórkowych modulatorów, mediatorów i transmiterów.Oba zostały zidentyfikowane w mnogości ścieżek metabolitycznych i procesów biochemicznych. Jednakże, przy braku stosownych mechanizmów kontroli, wymienione powyżej tak zwane „gazotransmitery” mogą wywoływać lub przynajmniej przyczyniać się do rozwoju wielu patofizjologii i zaburzeń, a nawet zagrażających życiu chorób. Najbardziej znamienne właściwości endogennych tlenku azotu i siarkowodoru bezpośrednio wynikają z ich chemicznych i fizycznych właściwości. Niepolarne gazotransmitery nie podlegają magazynowaniu w pęcherzykach, na wzór innych znanych neuroprzekaźników. Natychmiast po enzymatycznym zsyntetyzowaniu niemal nieograniczenie dyfundują przez cytozol oraz błony lipidowe do płynu zewnątrzkomórkowego i sąsiadujących komórek, wywierając efekt biologiczny na każdym etapie swojej drogi. W kontekście systemu nerwowego, gazotransmitery są jednymi z najistotniejszych nieklasycznych neuromodulatorów, zaangażowanych w szerokie spektrum procesów fizjologicznych i patofizjologicznych. Tlenek azotu i siarkowodór regulują między innymi tak ważne i istotne dla zachowania homeostazy układu nerwowego procesy jak plastyczność synaptyczna, potranslacyjna i funkcjonalna modyfikacja białek receptorowych na synapsach czy neuroprotekcja i neurodegeneracja.
It came as an epochal revelation as well as an incredible astonishment when commonly recognized noxious gaseous compounds - nitric oxide (NO) and hydrogen sulfide (H2S) - turned out to be vital and fundamental molecular elements of human metabolism and biochemistry. Acting under physiological conditions as intra- or intercellular modulators, facilitators and transmitters they have been identified in a multitude of metabolic pathways and biochemical processes. However, without proper control mechanisms, both so-called "gasotransmitters" mentioned above may cause or at least be involved in a plethora of pathophysiological statesand life-threatening diseases. The most striking features that identify endogenous nitric oxide and hydrogen sulfide are emerging directly from their chemical and physical properties. Gasotransmitters, due to their nonpolar chemical nature, are not stored in vesicles, like the rest of the neurotransmitters is. Immediately after enzymatic synthesis, they nearly unhinderedly start to diffuse in the cytosol and through cellular lipid membranes to the extracellular fluid as well as to the neighbour cells. This kind of signalling is blurred in comparison to classical synaptic transmission, although it is surprisingly potent and powerful. In the context of the nervous system, gasotransmitters are ones of the most prominent nonclassical neuromodulators and neurotransmitters. According to this, the main aim of this work is to describe a precisely selected array of the most appealing physiological and pathophysiological functions and tasks of gasotransmitters in the nervous system. Nitric oxide, for example, is involved in a phenomenon of synaptic plasticity, induction of long-term potentiation, regulation of sleep homeostasis and modulation of cell survival and degeneration. On the other hand, hydrogen sulfide is able to regulate extra- and intracellular levels of calcium and glutathione, to modify activation of membrane receptors and to arrange mechanisms of neuroprotection. Apart from these examples of functions, in this work, endogenous and exogenous sources of gasotransmitters in the environment of the nervous system will be revealed, as well as a brief history of the gaseous signalling molecules.
