dc.abstract.enPeriodontitis is a chronic inflammatory disease involving periodontal tissues that is caused by disturbances in the natural balance of the oral microbiome. The anaerobic bacterium Porphyromonas gingivalis belongs to the so-called "key pathogens" involved in this disease. Despite extensive knowledge about periodontitis, the contributions of epigenetic regulation to this disease are still not well understood. In many studies, differences in DNA methylation patterns between tissues from periodontitis patients and healthy donors have been reported. DNA methyltransferases (DNMT) are a family of enzymes responsible for DNA methylation. The aim of the study was to investigate the effect of DNMT inhibitors (5-azacytidine, deecitabine) on proliferation, metabolic activity, global DNA methylation, expression of inflammatory mediators and CCL20 production in primary human gingival fibroblasts (PHGF), and to investigate the effect of P. gingivalis ATCC 33277 infection on DNMT expression in these cells. DNMT inhibitors caused a decrease in proliferation while increasing the metabolic activity of PHGFs. They also caused global hypomethylation in PHGF. P. gingivalis-induced expression of CCL20, CCL2, CCL3, IL6 and IL8 was increased in cells treated with DNMT inhibitors. Further experiments involving PHGF stimulation with TNFα or IL 1β also showed an increase in the production of the CCL20 chemokine in cells treated with DNMT inhibitors. To investigate the effect of P. gingivalis on DNMT levels, the mRNA expression of DNMT1, DNMT3a and DNMT3b in cells infected with live and heat-inactivated bacteria was examined at two time points. After 4 hours of infection, there was a decrease in the expression of all examined DNMTs, whereas after 24 hours there was an increase in the expression of DNMT1 in all tested conditions and of DNMT3b after infection with live bacteria. However, Western blot analysis showed a decrease in DNMT1 protein levels in PHGFs after infection with a high MOI of bacteria (MOI 100: 1). In conclusion, the results suggest that P. gingivalis may induce hypomethylation in PHGFs by lowering the protein level of DNMT1, which may result in increased expression of inflammatory mediators.pl

Pomimo dużej wiedzy na temat zapalenia przyzębia, mechanizmy regulacji epigenetycznej w tej chorobie wciąż nie są dobrze poznane. W wielu badaniach stwierdzono różnicę w metylacji DNA między tkankami od pacjentów z paradontozą, a tymi od zdrowych dawców. Enzymami odpowiadającymi za metylację DNA są metylotransferazy DNA (DNMT). Celem pracy było zbadanie wpływu inhibitorów DNMT (5-azacytydyna, deecytabina) na proliferacje, aktywność metaboliczną, globalną metylację oraz ekspresję mediatorów stanu zapalnego i produkcje CCL20 w pierwotnych ludzkich fibroblastach dziąsła (PHGF) oraz scharakteryzowanie wpływu infekcji bakteriami P. gingivalis ATCC 33277 na ekspresję DNMT w tych komórkach. Inhibitory DNMT w komórkach PHGF powodowały spadek proliferacji przy jednoczesnym podniesieniu aktywności metabolicznej tych komórek. Wywołały one również globalną hipometylację w PHGF. W komórkach traktowanych inhibitorami DNMT zaobserwowano podwyższenie ekspresji CCL20, CCL2, CCL3, IL6 i IL8 indukowanej przez infekcję P. gingivalis. Dalsze doświadczenie polegające na stymulacji komórek przy użyciu TNFα lub IL-1β również pokazały wzrost produkcji chemokiny CCL20 w komórkach traktowanych inhibitorami DNMT. W celu zbadania wpływu P. gingivalis na DNMT, zbadano poziom ekspresji mRNA dla DNMT1, DNMT3a i DNMT3b w komórkach infekowanych żywymi i inaktywowanymi cieplnie bakteriami, w dwóch punktach czasowych. Po 4 godzinach infekcji nastąpił spadek ekspresji wszystkich badanych DNMT, natomiast po 24 godzinach nastąpił wzrost ekspresji DNMT1 w każdych warunkach infekcji oraz DNMT3b tylko dla infekcji żywymi bakteriami. Jednak analiza Western blot pokazała spadek ekspresji białka DNMT1 w komórkach PHGF po infekcji bakteriami o wysokim MOI (MOI 100:1), co może być spowodowane degradacją DNMT1 przez proteazy bakteryjne. Podsumowując, otrzymane wyniki sugerują, że P. gingivalis może wywoływać hipometylację w komórkach PHGF poprzez obniżenie poziomu DNMT1, co może skutkować zwiększeniem ekspresji mediatorów stanu zapalnego.pl
