Rola receptorów adenozynowych A2A w efektach behawioralnych kokainy u szczurów

dc.abstract.enCocaine addiction is a serious socio-medical problem all over the world. Despite the well-known mechanism of cocaine action, which depends of the ability of the drug to block dopamine, serotonin and noradrenalin reuptake. Presently, there is no medication approved for the treatment of cocaine addiction. Despite the high affinity of cocaine to dopamine transporter and dopamine D2 receptor, and changes in dopamine neurotransmission, treatment of cocaine addiction is focused on dopaminergic neuro-transmission. It is postulated that adenosine A2A receptors may play a significant role in the treatment of cocaine addiction. It is related with the existence of antagonistic interaction between A2A-D2 receptors. The stimulation of A2A receptors produces some kind of steric modification of D2 receptors, with the change in its binding properties. It was postulated that this A2A-D2 receptor interaction requires a close proximity between both receptors, with the possible existence of heterometric receptor complex.The aim of these study was to compare the effects of A2A receptor ligands (a selective agonist and antagonist) on rewarding properties of artificial reward, that is cocaine, in a model of cocaine self-administration in rats. In the experiment, selective A2A receptor ago-nist CGS 21680, and selective A2A receptor antagonist SCH 58261 were used.The study demonstrated, that A2A receptor agonist CGS 21680 did not attenuate active and inactive lever presses, however, it decreased the number of cocaine infusions during cocaine self-administration. The selective A2A receptor antagonist SCH 58261 did not in-fluence on cocaine rewarding properties.In the reinstatement phase induced by cocaine (10 mg/kg, ip), the A2A receptor agonist CGS 21680 prevented the reinstatement of cocaine seeking behavior seen as reduction in the number of active lever presses. The A2A receptor antagonist SCH 58261 caused the reinstatement of cocaine seeking behavior by increasing the number of active lever presses, without the influence on inactive lever. These findings suggest that pharmacological stimulation of A2A receptors counteracts against rewarding properties of artificial reward.pl

Pomimo dobrze poznanego mechanizmu działania kokainy, związanego z bloko-waniem wychwytu zwrotnego monoamin – dopaminy, serotoniny i noradrenaliny, brak jest obecnie skutecznej farmakoterapii tej choroby. Z uwagi na powinowactwo kokainy do DAT i receptorów D2 oraz zmian w neurotransmisji dopaminowej, leczenie uzależnienia kokainowego obejmuje głównie wpływ na ten układ. W ostatnich latach postuluje się, że receptory A2A mogą odgrywać istotną rolę w leczeniu nałogu kokainowego. Związane jest to z istnieniem antagonistycznej interakcji pomiędzy receptorami D2, a receptorami A2A w strukturach mózgu warunkujących fenotyp uzależnienia. I tak, stymulacja receptorów A2A wywołuje modyfikacje w funkcji receptorów D2 (np. zmniejszenie ich zdolności do wiązania się z agonistami). Interakcja ta wymaga ich bliskiego położenia względem siebie, w postaci tak zwanych kompleksów heteromerycznych. Celem niniejszej pracy było zbadanie wpływu ligandów receptorów A2A na efekty na-gradzające sztucznej nagrody jaką jest kokaina, w modelu dożylnego samopodawania ko-kainy u szczurów. Badanym szczurom podawano selektywnego agonistę receptorów A2A CGS 21680, oraz selektywnego antagonistę tych receptorów SCH 58261. W fazie podtrzymywania samopodawania kokainy wykazano, że agonista receptorów A2A – CGS 21680 nie wpłynął na liczbę naciśnięć na dźwignię aktywną, jednakże zmniej-szył liczbę infuzji kokainowych. Selektywny antagonista SCH 58261 nie wpłynął na liczbę naciśnięć na dźwignie aktywną oraz nieaktywną, ani nie zmieniał liczby infuzji kokai-nowych.W fazie nawrotów, indukowanych podaniem kokainy (10 mg/kg, ip) agonista recepto-rów A2A CGS 21680 spowodował zahamowanie nawrotu zachowań poszukiwawczych kokainy, zmniejszając liczbę naciśnięć na dźwignię aktywną. Antagonista receptorów A2A SCH 58261 wywołał nawrót zachowań poszukiwawczych, zwiększając liczbę naciśnięć na dźwignię aktywną, przy braku wpływu na dźwignię nieaktywną.Z przeprowadzonych badań wynika, iż farmakologiczna stymulacja receptorów A2A przeciwdziała właściwościom nagradzającym sztucznej nagrody.pl
