Powiązanie pomp protonowych i fototropin w przekazie sygnałów sterujących ruchami chloroplastów
The role of proton pumps and phototropins in signalling associated with chloroplast movements.

Prawidłowe stężenie protonów wewnątrz komórki roślinnej jest utrzymywane przez plazmolemowe pompy protonowe (H+-ATPazy). Transport protonów na zewnątrz komórki tworzy gradient elektrochemiczny, który wykorzystywany jest w podstawowych procesach fizjologicznych (regulacja pH, transport jonów i metabolitów) oraz w odpowiedzi na warunki stresowe (np. tolerancja na zasolenie). Arabidopsis thaliana posiada 11 genów kodujących H+-ATPazy – AHA1-11. H+-ATPazy biorą udział w ścieżkach przekazu sygnału w procesach regulowanych światłem niebieskim: ruchu aparatów szparkowych, ruchu liścia oraz fototropizmie. Możliwe jest więc zaangażowanie pomp protonowych w inne procesy kontrolowane przez fototropiny. Jednym z sugerowanych procesów jest ruch chloroplastów – użycie inhibitorów H+-ATPazy spowodowało zahamowanie odpowiedzi ruchowej chloroplastów na światło niebieskie. Przeprowadzone w pracy badania miały na celu sprawdzenie zaangażowania pomp protonowych w przekaz sygnałów świetlnych do ruchów chloroplastów. Wykorzystano mutanty insercyjne T-DNA genów aha3 oraz aha11. Użycie mutantów pomp, a nie inhibitorów, zapewniło brak funkcjonalnej pompy w roślinie oraz wyeliminowało ryzyko niespecyficznego działania inhibitora. Ruch chloroplastów mierzono metodą fotometryczną. Wyniki badań nie wykazały udziału badanych izoform H+-ATPaz w ruchu chloroplastów. Ze względu na możliwe występowanie zjawiska redundancji funkcjonalnej, otrzymane wyniki nie dają pewności o braku zaangażowania H+-ATPaz w przekaz sygnałów świetlnych do ruchów chloroplastów.Ze względu na zaangażowanie pomp protonowych w wielu procesach fizjologicznych roślin oraz w dostosowaniu się do niekorzystnych warunków środowiska, H+-ATPazy stanowią atrakcyjny cel badań biotechnologii.
Concentration of protons inside plant cells is controlled by plasma membrane proton pumps (H+-ATPases). Transport of protons out of the cell creates an electrochemical gradient used in basic physiological processes such as pH regulation or transport of ions and metabolites. Proton gradients are also important during response to stress conditions such as salt stress. Arabidopsis thaliana has 11 genes encoding H+-ATPases - AHA1-11. H+-ATPases are involved in signal transduction pathways in blue light-regulated processes: stomata opening, leaf movement and phototropism. It is possible that proton pumps are involved in other processes controlled by phototropins. One of the suggested processes, that may be influenced by the activity of proton pumps, is the movement of chloroplasts. This hypothesis is supported by the fact that H+-ATPase inhibitors have been shown to inhibit the blue light driven chloroplast relocation. Research conducted in this thesis aimed to establish the involvement of proton pumps in the transmission of light signals in chloroplast movements. The A. thaliana T-DNA insertion mutants of the aha3 and aha11 genes were used. The use of mutants, not inhibitors, ensured a specific lack of pump in the plant cell and eliminated the risk of inhibitor nonspecificity. The movement of chloroplasts was measured using the photometric assay. The results do not suggest any significant contribution of AHA3 and AHA11 H+-ATPase in the control of the chloroplast movement. Since the pumps encoded by the AHA gene family are likely to be functionally redundant, the obtained results do not completely exclude the involvement of H+-ATPases in the transmission of light signals in signalling associated with chloroplast movements. Due to the involvement of proton pumps in many plant physiological processes and their role in plant adaptation to adverse environmental conditions, H+-ATPases are an attractive target for biotechnology research.
