Integralność DNA plemników stanowi jeden z kluczowych wyznaczników jakości nasienia, determinując płodność męską oraz prawidłowy przebieg wczesnych etapów rozwoju zarodkowego. Pomimo rosnącego znaczenia tego parametru w diagnostyce andrologicznej, jego ocena nadal opiera się głównie na metodach pośrednich, takich jak test struktury chromatyny plemników (Sperm Chromatin Structure Assay, SCSA), który bazuje na analizie podatności DNA na denaturację, a nie na bezpośrednim pomiarze uszkodzeń. Celem niniejszej pracy była ocena przydatności metody L1-LORD-Q (L1-long-run real-time PCR based DNA-damage quantification) jako narzędzia umożliwiającego bezpośrednią, ilościową analizę integralności DNA plemników oraz jej porównanie z metodą SCSA z wykorzystaniem modelu mysiego. Badania przeprowadzono w dwóch etapach eksperymentalnych, obejmujących zarówno plemniki z indukowanymi nadtlenkiem wodoru uszkodzeniami DNA, jak i próbki o naturalnym poziomie fragmentacji, które oceniano również pod kątem podstawowych parametrów jakości nasienia. Uzyskane wyniki wykazały, że obie metody umożliwiają detekcję wzrostu poziomu uszkodzeń DNA w warunkach indukowanego stresu oksydacyjnego, jednak pomiary wykonane metodą L1-LORD-Q charakteryzują się większą liniowością i zdolnościami rozdzielczymi nawet w najwyższych dawkach nadtlenku wodoru. W przypadku próbek o naturalnym poziomie fragmentacji zgodność wyników obu metod była ograniczona. Analiza korelacji z innymi parametrami jakości plemników wykazała, że wyniki uzyskane metodą L1-LORD-Q zdecydowanie lepiej niż SCSA odzwierciedlają rzeczywisty stan nasienia. Przeprowadzone badania sugerują, że metoda L1-LORD-Q stanowi obiecujące narzędzie w ocenie integralności DNA plemników, mogące pełnić rolę uzupełniającą, a potencjalnie również alternatywną wobec metod opartych na analizie pośredniej. Jej zastosowanie może przyczynić się do zwiększenia precyzji oceny jakości nasienia oraz poprawy diagnostyki niepłodności męskiej.
Sperm DNA integrity is one of the key determinants of semen quality, affecting male fertility as well as the proper progression of early embryonic development. Despite the growing importance of this parameter in andrological diagnostics, its assessment still relies largely on indirect methods, such as the sperm chromatin structure assay (SCSA), which is based on the evaluation of DNA vulnerability to denaturation rather than on the direct measurement of DNA damage. The aim of this study was to evaluate the applicability of the L1-LORD-Q (L1-long-run real-time PCR-based DNA-damage quantification) method as a tool enabling direct, quantitative analysis of sperm DNA integrity and to compare it with the SCSA method using a mouse model. The study was conducted in two experimental stages, including both sperm samples with hydrogen peroxide-induced DNA damage and samples with natural levels of DNA fragmentation, which were additionally evaluated for basic semen quality parameters. The obtained results demonstrated that both methods allow for the detection of increased DNA damage under conditions of induced oxidative stress; however, measurements performed using the L1-LORD-Q showed a more proportional increase and greater discriminative capacity, even at the highest concentrations of hydrogen peroxide. In the case of samples with natural levels of DNA fragmentation, the agreement between the two methods was limited. Correlation analysis with other sperm quality parameters revealed that the results obtained using the L1-LORD-Q method better reflect the actual state of semen compared to SCSA. These findings suggest that the L1-LORD-Q method is a promising tool for the assessment of sperm DNA integrity, potentially serving as a complementary or even alternative approach to methods based on indirect analysis. Its application may contribute to improving the precision of semen quality evaluation and enhancing the diagnostics of male infertility.
