Wpływ aktywności RNazowej białka MCPIP1 na szlak degradacji β-kateniny w linii komórkowej Caki-1
The influence of the RNase activity of the MCPIP1 protein on the β-catenin degradation pathway in the Caki-1 cell line

β-katenina odgrywa kluczową rolę w regulacji kanoniczej ścieżki szlaku Wnt regulując ważne procesy komórkowe m.in różnicowanie, proliferację komórek oraz embriogenezę. Wiele obecnych badań dowodzi silnego związku między szlakiem Wnt/β-katenina a różnymi aspektami nowotworzenia, w tym inicjacją, przerzutowaniem, lekoopornością oraz utrzymaniem cech macierzystych komórek nowotworowych. W jasnokomórkowym raku nerki wraz ze wzrostem β-kateniny obserwuje się spadek poziomu białka MCPIP1, regulującego rozwój tego nowotworu. MCPIP1 dzięki aktywności RNazowej hamuje szlak Wnt przez dezaktywację β-kateniny. Celem badań było określenie roli aktywności RNazowej białka MCPIP1 na szlak degradacji β-kateniny. W przedstawionej pracy analizowano poziom wybranych form modyfikowanej potranslacyjnie β-kateniny oraz zmiany lokalizacji β-kateniny po zastosowaniu inhibitora proteasomu MG132. Doświadczenia przeprowadzono w linii komórkowej jasnokomórkowego raka nerki, Caki-1, ze stabilną nadekspresją zmutowanej oraz dzikiej formy białka MCPIP1. Otrzymane wyniki wykazały wzrost akumulacji β-kateniny po zastosowaniu inhibitora MG132 niezależnie od aktywności RNazowej białka MCPIP1, w tym głównie β-kateniny fosforylowanej na resztach Ser33/Ser37/Thr41. Podsumowując, w linii komórkowej Caki-1 ze zmutowaną oraz dziką formą białka MCPIP1 mechanizm regulacji aktywnej β-kateniny wydaje się być niezależny od klasycznej ścieżki degradacji w proteasomie.
β-catenin plays a key role in regulating the canonical Wnt pathway, controlling important cell processes such as differentiation, cell proliferation and embryogenesis. Many current studies demonstrate a strong association between Wnt/β-catenin pathway and various aspects of carcinogenesis including initiation, metastasis, drugs resistance and the maintenance of cancer stem cells properties. In clear cell renal cell carcinoma (ccRCC), an increase in β-catenin is observed along with a decrease in the level of MCPIP1 protein, which regulates the development of this cancer. MCPIP through its RNase activity inhibits the Wnt pathway by deactivation of β-catenin. The aim of this study was to determine the role of RNase activity of MCPIP1 in β-catenin degradation pathway. The presented study analysed the levels of selected post-translationally modified forms of β-catenin and changes in β-catenin location after treatment with the proteasome inhibitor MG132. Experiments were conducted on ccRCC cell line, with stable overexpression of both mutated and wild type MCPIP1. The obtained results showed an increase in β-catenin accumulation after MG132 treatment, regardless RNase activity of MCPIP1, mainly in β-catenin phosphorylated at Ser33/Ser37/Thr41. Concluding, in Caki-1 with mutated and wild type MCPIP1, the regulation mechanism of active form of β-catenin appears to be independent of classical degradation pathway in proteasome.
