The influence of heme metabolism on endothelial cell senescence and quiescence

Hem jest niezbędnym kofaktorem zaangażowanym w wiele kluczowych procesów biologicznych. Nie mniej, poziom hemu musi być ściśle regulowany by zapewnić homeostazę. Kluczowe wydaje się uzupełnienie wiedzy na temat roli wolnego hemu w śródbłonku naczyniowym, w szczególności w kontekście wpływu dysfunkcji śródbłonka na rozwój wielu chorób. W niniejszej pracy skupiono się na analizie wpływu metabolizmu hemu w komórkach śródbłonka na starzenie się i stan uśpienia pierwotnych ludzkich komórek śródbłonka aorty. Stymulacja hemem lub obniżona synteza hemu nie wpłynęły na cykl komórkowy, nie mniej, stymulacja heminą zwiększała liczbę komórek starzejących się. Co ciekawe, stymulacja δALA, prekursorem hemu, nie powodowała takiego efektu. Zwiększenie wolnego hemu przez stymulację δALA nie zmieniła ekspresji genów związanych ze stanem uśpienia w komórkach śródbłonka. Zaprezentowane wyniki pozwalają lepiej zrozumieć wpływ wolnego hemu na starzenie i uśpienie komórek śródbłonka i mogą służyć jako dane wyjściowe do dalszych badań.
Heme is an essential cofactor involved in various crucial biological processes within cells. However, its levels should be tightly regulated in order to maintain cellular homeostasis. To date, little is known about the influence of heme in vascular endothelial cells, and considering the contribution of dysfunctional endothelial cells to the development and progression of multiple diseases, it seems of great importance to address this gap of knowledge. Therefore, the present study aimed to evaluate the influence of heme metabolism on endothelial cell senescence and quiescence. Although heme surplus or heme deficiency did not affect cell cycle progression, heme surplus by hemin resulted in increased senescence of endothelial cells, an interesting pattern not observed after δALA supplementation. Furthermore, we found that heme excess by δALA did not result in the overexpression of quiescence-related genes and possibly does not lead to a pro-quiescent endothelial cell phenotype as previously thought. Altogether, these data provide valuable insights into the influence of heme on endothelial cell metabolism and serve as the starting point for future experiments that will shed light on this relationship.
