Synteza nowych ligandów wielofunkcyjnych wpływających na procesy neurodegeneracyjne

dc.abstract.enThe aim of the study was the synthesis of novel multi-target-directed ligands (MTDLs), which can affect the neurodegenerative processes involved in Alzheimer's disease. The synthesized compounds were designed by molecular modeling. The introduction to this work contains a brief overview of idea of multi-target-directed ligands (MTDLs), selected examples of their design strategies and exemplary MTDLs, designed as potential anti-AD drug candidates. Author’s own research involved a synthesis of a new series of compounds with a potential cholinesterase and beta-secretase inhibitory activity. In the first step of the synthesis, the appropriate substrates – bromoalkyl derivatives of saccharin and phthalimide and arylalkyl derivatives of piperazine and 3-aminopiperidine were obtained. They were used for the synthesis of the final compounds in the next step. The identity of all the synthesized compounds was confirmed by LC-MS, 1H NMR and, in the case of the final compounds, by 13C NMR. Overall, fourteen final compounds were synthesized and then tested in the Ellman’s assay, in order to determine their inhibitory activity against acetyl- (AChE) and butyrylcholinesterase (BuChE). At the screening concentration of 10^-5 M, three of the examined compounds, derivatives of saccharin containing the 1-(2-fluorobenzyl)piperazine fragment, inhibited AChE in more than 50%. The determined IC50 values for these compounds ranged from 4,25 μM to 17,80 μM. One of the tested compounds showed activity against BuChE. The IC50 value determined for this compound was 0,33 μM, which allows to classify this derivative as a potent inhibitor of BuChE.pl

Związki te zostały zaprojektowane metodami modelowania molekularnego. Wstęp do niniejszej pracy prezentuje ideę ligandów wielofunkcyjnych, wybrane strategie ich projektowania, a także przykłady takich związków, zaprojektowanych jako potencjalne leki na AD. W ramach badań własnych przeprowadzono syntezę związków o potencjalnym działaniu hamującym cholinoesterazy i beta-sekretazę. W pierwszym etapie otrzymano odpowiednie substraty - bromoalkilowe pochodne sacharyny i ftalimidu oraz aryloalkilowe pochodne piperazyny i 3-aminopiperydyny. Związki te w kolejnym etapie posłużyły do syntezy związków finalnych. Tożsamość wszystkich otrzymanych pochodnych potwierdzono za pomocą analizy LC-MS, 1H NMR, oraz, w przypadku związków finalnych, 13C NMR. W sumie otrzymano 14 związków finalnych, które poddano badaniu aktywności wobec acetylocholinoesterazy (AChE) oraz butyrylocholinoesterazy (BuChE), stosując spektrofotometryczny test Ellmana. W stężeniu przesiewowym (10^-5 M), trzy z przebadanych związków, pochodne sacharyny zawierające fragment 1-(2-fluorobenzylo)piperazyny, hamowały AChE w ponad 50%. Wyznaczone dla nich wartości IC50 wynosiły od 4,25 μM do 17,80 μM. Jedna z przebadanych pochodnych była aktywna wobec BuChE. Uzyskana wartość IC50 dla tego związku wyniosła 0,33 μM, co pozwala na uznanie tej pochodnej za silny inhibitor BuChE.pl
