Wpływ inhibitora SU11274 na poziom fosforylacji receptora Met i poziom białka MCPIP1 w linii komórkowej Caki-2

Receptorowa kinaza tyrozynowa Met aktywowana przez ligand, czynnik wzrostu hepatocytów (HGF) , może być znacząco związany z progresją jasnokomórkowego raka nerki (ccRCC). W niniejszym badaniu, stosując linię komórkową Caki-2 wykazano że inhibitor Met, SU11274 hamował fosforylację receptora zmniejszając tym samym poziom aktywnego receptora w komórkach. Hamowanie szlaku Met-HGF może prowadzić do zahamowania angiogenezy guza i zmniejszenia żywotności jasnokomórkowego raka nerki. Ponadto w doświadczeniu zbadano ekspresję białka MCPIP1, które również wpływa na rozwój ccRCC. MCPIP1 pośredniczy w procesach zapalnych poprzez hamowanie ekspresji cytokin prozapalnych oraz kontrolowanie aktywności czynników transkrypcyjnych NF-γB i AP1. Wyniki doświadczenia wykazały wzrost poziomu MCPIP1 podczas hamowania receptora Met. Jak wykazano w badaniach, MCPIP1 prowadzi do zmniejszenia żywotności komórek poprzez jednoczesne indukowanie apoptozy w niektórych typach raka. Dlatego receptor Met wydaje się być dobrym celem terapii nowotworowej. Hamowanie receptora Met może przyczynić się do ograniczenia wzrostu jasnokomórkowego raka nerki.
Met tyrosine kinase receptor activated by ligand, hepatocyte growth factor (HGF), may be significantly associated with the progression of clear cell renal cell carcinoma (ccRCC). In the present study, using cell line Caki-2, it was demonstrated that the c-Met inhibitor SU11274 inhibited receptor phosphorylation thereby reducing the level of active receptor in cells. Inhibition of the Met-HGF pathway may lead to suppression of tumor angiogenesis and decrease in viability of clear cell renal cell carcinoma. Furthermore, the experiment investigated the expression of MCPIP1 protein, which also participates in the development of ccRCC. MCPIP1 mediates inflammatory processes by inhibiting the expression of proinflammatory cytokines, and controlling activity of NF-γB and AP1 transcription factors. Results of experiment showed an increase in MCPIP1 levels during MET receptor inhibition. As shown by studies, MCPIP1 leads to decreased cell viability by simultaneous induction of apoptosis in certain types of cancer. That is why the Met receptor seems to be a good target for cancer therapy. Inhibition of the Met receptor may contribute to limiting the growth of renal cell carcinoma.
