Dysfunkcje tarczycy, takie jak nadczynność i niedoczynność, mogą wpływać na rozwój nowotworów jajnika poprzez oddziaływanie na metabolizm organizmu. W kontekście złożonego wpływu różnych czynników na rozwój raka jajnika, hormony tarczycy odgrywają kluczową rolę, regulując wiele procesów biologicznych w jajniku. Wśród nowotworów jajnika wyróżniane są rzadko występujące i słabo poznane ziarniszczaki jajnika. Dojrzały typ ziarniszczaka jajnika cechuje się późnymi nawrotami choroby bez możliwości ich wyleczenia. Dlatego celem niniejszej pracy było określenie wpływu hormonu T3 (w dawce występującej we krwi u pacjentek ze zdiagnozowaną nadczynnością tarczycy) na metabolizm komórek ziarniszczaka jajnika (KGN), co może dostarczyć nowych informacji na temat mechanizmów leżących u podstaw tego nowotworu. Kontrolę do badań stanowiła linia prawidłowa komórek ziarnistych jajnika (HGrC1). Poziom glikolizy (glycoATP) oraz fosforylacji oksydacyjnej (mitoATP) w komórkach oceniano poprzez równoczesny pomiar OCR (ang. oxygen consumption rate) i ECAR (ang. extracellular acidification) w czasie rzeczwistym, za pomocą testu Agilent Seahorse XFp Real-Time ATP Rate Assay Kit z zastosowaniem urządzenia Seahorse XFp. Wyniki naszych badań wykazały różnicę w bazalnym poziomie metabolicznym komórek HGrC1 i KGN. Co więcej wykazano, że poziom mitoATP w komórkach prawidłowych HGrC1, po stymulacji T3, był wyższy niż w komórkach prawidłowych bez stymulacji T3. W przypadku komórek nowotworowych (KGN) stymulacja hormonem T3 nie wpłynęła istotnie na produkcję mitoATP oraz glycoATP. Podsumowując, nasze badania dostarczają nowych danych na temat wpływu T3 na poziom metabolizmu komórek ziarnistych jajnika. Zwiększenie metabolizmu prawidłowych komórek ziarnistych jajnika pod wpływem T3 wskazuje na ważną rolę tego hormonu w regulacji funkcji jajnika oraz w zrozumieniu mechanizmów wpływających na rozwój raka jajnika.
Thyroid dysfunctions, such as hyperthyroidism and hypothyroidism, can influence the development of ovarian cancers by affecting the body's metabolism. Given the complex influence of several factors on the development of ovarian cancer, thyroid hormones play a key role by regulating many biological processes in the ovary. Among ovarian tumours, granulosa cell tumours (GCTs) are rarely occurring and poorly understood. The mature type of granulosa cell tumour is characterised by late recurrences without the possibility of cure. Given the complex influence of several factors on the development of ovarian cancer, thyroid hormones play a key role by regulating many biological processes in the body, including in the ovary. Therefore, the aim of this study was to prove the effect of the T3 hormone (at a dose found in the blood of patients diagnosed with hyperthyroidism) on the metabolism of granulosa cell tumour cells (KGN), which may offer new insights into the mechanisms underlying this cancer. The control for the study was a normal ovarian granulosa cell line (HGrC1). The levels of glycolysis (glycoATP) and oxidative phosphorylation (mitoATP) in the cells were measured by simultaneous measurement of OCR (oxygen consumption rate) and ECAR (extracellular acidification) in real-time using the Agilent Seahorse XFp Real-Time ATP Rate Assay Kit with the Seahorse XFp device. Our results show differences in the basal metabolic levels of HGrC1 and KGN cells. Moreover, it has been shown that the level of mitoATP in normal HGrC1 cells, after T3 stimulation, was higher than in normal cells without T3 stimulation. In the case of cancer cells (KGN), T3 stimulation did not significantly affect their mitoATP and glycoATP production.In summary, our research provides new data on the impact of T3 on the metabolic level of ovarian granulosa cells. The increase in metabolism of normal ovarian granulosa cells under the influence of T3 indicates the key role of this hormone in regulating ovarian function as well as in understanding the mechanisms influencing ovarian cancer development.
