Wykrywanie i wpływ mikroplastiku na żywotność komórek w kulturach zawiesinowych Arabidopsis arenosa (L.) Lawalrée

Zanieczyszczenie mikroplastikiem stanowi poważne zagrożenie dla środowiska, które jest obecnie jednym z największych problemów ekologicznych współczesnego świata. Jego wpływ na rośliny nie jest jednak dokładnie zbadany. Celem niniejszej pracy było wykrycie i zbadanie wpływu trzech rodzajów mikroplastiku (MP) o wielkości cząsteczek 0,1 μm na kultury zawiesinowe komórek Arabidopsis arenosa, gatunku tolerującego suszę i akumulującego metale ciężkie. Wyniki badań wykazały, że komórki A. arenosa akumulują zarówno polistyren zmodyfikowany grupą aminową, siarczanową, jak również polistyren niezmodyfikowany w obu stężeniach (100 μg/L oraz 500 μg/L) oraz że zakumulowany plastik nie wpływa negatywnie na żywotność komórek niezależnie od czasu i dawki traktowania. Co więcej, po 72h traktowania polistyrenem zmodyfikowanym grupą siarczanową żywotność komórek zwiększyła się o ok. 95% w stosunku do kontroli (0 μg/L) w obu zastosowanych stężeniach. Wydaje się, że stosunkowo niewielkie stężenie zastosowanego MP wywołało hormezę (pozytywny efekt niewielkiej ilości czynnika, w dużej dawce wywołującego negatywny efekt). Badania nadwpływem MP na kultury zawiesinowe są całkiem nowym obszarem badawczym. Aby móc stwierdzić jednoznaczny efekt wpływu różnych grup funkcyjnych mikroplastiku, potrzeba więcej szczegółowych badań porównujących wpływ danych cząsteczek na komórki konkretnych gatunków roślin. W perspektywie badań, powinny się znaleźć eksperymenty sprawdzające dotychczas słabo poznane interakcje między MP a metalami ciężkimi, a A. arenosa, ze względu na swoją tolerancję na metale ciężkie może być do tego dobrym modelem badawczym.
Microplastic pollution poses a serious threat to the environment, which is currently one of the biggest ecological problems in the modern world. However, its effect on plants has not been thoroughly investigated. The aim of this study was to detect and examine three types of microplastics (MP) with a particle size of 0.1 μm on suspension cultures of Arabidopsis arenosa cells, a drought-tolerant species that accumulates heavy metals. The results showed that A. arenosa cells accumulate both polystyrene modified with an amino and sulfate group, as well as unmodified polystyrene at both concentrations (100 μg/L and 500 μg/L) and that the accumulated plastic does not negatively affect cell viability regardless of time and treatment dose. Moreover, after 72 h of treatment with sulfate-modified polystyrene, cell viability increased by approximately 95% compared to the control (0 μg/L) at both applied concentrations. It seems that the relatively low concentration of MP used had a hormesis effect on the cells (a positive effect of a small amount of a factor causing a negative effect in a large dose). Research on the impact of MP on suspension cultures is a completely new research area. To be able to state a clear effect of the influence of various functional groups of microplastics, more detailed research is needed to compare the influence of given molecules on the cells of specific plant species. In the research perspective, there should be experiments checking the hitherto poorly understood interactions between MP and heavy metals, and A. arenosa, due to its tolerance to heavy metals, may be a good research model for this purpose.
