dc.abstract.enMajor depression is a recurrent and debilitating mental disorder with a lifetime prevalence of up to 20% in the general population, among the highest for psychiatric disorders. The aetiology is still largely unknown, and the molecular and cellular mechanism underlying depression and the actions of antidepressant treatment have not been fully elucidated. It was found that chemical antidepressant and electroconvulsive seizures induce neural proliferation in progenitor cells. Given that stress is thought to precipitate and exacerbate depression, the regulation of SGZ neurogenesis by stress has been extensively studied in a variety of species. Adult neurogenesis is a dynamic and multistage process that is regulated by physiological, pathological and pharmacological factors. The potential mechanism may be very diverse (changes in neurotransmitters, neurotrophic factors, intracellular cascades and genes). The effects of antidepressants on all stages of neurogenesis (proliferation, differentiation, and survival of newborn neurons in dentate gyrus) have been proved. The behavioral effects of antidepressants are mediated by both neurogenesis-dependent and neurogenesis-independent mechanisms. Recent studies on newborn neurons show that these cells may function as modulators of principal neurons to influence pattern separation and that adult neurogenesis constitutes an adaptive mechanism to optimally encode contextual information. Based on current evidence, adult hippocampal neurogenesis is more likely a substrate for some of behavioral effects of antidepressants than a pivotal contributor to the etiology of depression.pl
dc.abstract.plDepresja jest chroniczną dewastującą chorobą, dotykającą około 20% populacji, której etologia i mechanizmy odpowiedzi na leczenie ciągle nie są znane. Gdy odkryto, że chemiczne antydepresanty jak i elektrowstrząsy stymulują proliferację neuralnych progenitorów, a stres, który jest obecnie uważany za jedną z głównych przyczyn depresji, ją obniża, zaczęto intensywnie badać wpływ terapii antydepresyjnych na neurogenezę. Neurogeneza u dorosłych jest wielostopniowym i dynamicznym procesem, który poddawany jest ścisłej regulacji przez czynniki fizjologiczne, ale i patologiczne oraz farmakologiczne. Mało tego, regulowana jest na różnych etapach, od zmian w neuroprzekaźnikach, poprzez czynniki neurotroficzne, do ścieżek sygnalizacyjnych i genów. Jak udowodniono, antydepresanty wpływają zarówno na proliferację, jak i dojrzewanie oraz przeżycie nowopowstałych neuronów w zakręcie zębatym. Z kolei wpływ neurogenezy na antydepresyjne efekty behawioralne nie jest już taki jednoznaczny. Niespójność wyników próbuje się tłumaczyć różnicami w testach behawioralnych, szczepie zwierząt oraz tym, czy były wcześniej stresowane. Ostatnie badania na nowopowstałych neuronach w zakręcie zębatym u dorosłych zwróciły uwagę na ich rolę w rozróżnianiu wzorców i ich generalizacji. Świadczy to o funkcjonalnym aspekcie nowopowstałych neuronów, a zaburzony stosunek między tymi dwoma zjawiskami wydaje się mieć konsekwencje m.in. w zaburzeniach afektywnych. Dotychczasowe wyniki wskazują, że depresja wydaje się mieć przynajmniej częściowy związek z dorosłą neurogenezą.pl
