Ocena poziomu transkryptów dla elongazy 5 i desaturazy 6 w tkance tłuszczowej szczurów otrzymujących fluwastatynę.
Evaluation of transcript levels for elongase 5 and desaturase 6 in adipose tissue of rats receiving fluvastatin.

Statyny, w tym fluwastatyna, będące inhibitorami reduktazy HMG-CoA, są stosowane powszechnie jako leki hipolipemizujące. Przypuszcza się, że statyny, mogą wpływać także na endogenną syntezę wielonienasyconych kwasów tłuszczowych, w której kluczową rolę pełnią elongazy i desaturazy kwasów tłuszczowych. Stwierdzono, ze fluwastatyna istotnie statystycznie obniża poziom mRNA dla Δ6D, natomiast nie wpływa na poziom mRNA dla Elovl-5 w tkance wątrobowej szczurów. Do tej pory nie przeprowadzano badań mających na celu ocenę działania fluwastatyny na kluczowe enzymy w syntezie PUFA w tkance tłuszczowej. Celem niniejszej pracy była ocena działania fluwastatyny na poziom mRNA dla Elovl-5 i Δ6D w tkance tłuszczowej szczurów. W przebiegu doświadczenia wykorzystano tkanki tłuszczowe szczurów płci męskiej rasy Wistar, które pochodziły z poprzednio wykonywanych badań w Zakładzie Analityki Biochemicznej UJ CM. Grupie badanej (n=6) za pomocą sondy dożołądkowej podawano przez 7 kolejnych dni fluwastatynę w dawce 25 mg/kg m.c./dobę, a grupie kontrolnej (n=5) wodę, będącą rozpuszczalnikiem leku. Ocenę poziomu mRNA dla Elovl-5 i Δ6D w tkance tłuszczowej wykonano przy pomocy półilościowej metody RT-PCR (odwrotna transkrypcja-PCR). Chociaż średni poziom mRNA dla Elovl-5 w grupie badanej był wyższy, zaś poziom mRNA dla Δ6D był niższy niż w grupie kontrolnej, to jednak różnice te nie były istotne statystycznie. Można zatem twierdzić, że fluwastatyna w dawce 25 mg/kg m.c nie wpływa na poziom mRNA dla tych enzymów w tkance tłuszczowej. Przyczyną różnic tkankowych działania fluwastatyny na poziom mRNA dla Δ6D można wytłumaczyć sposobem dystrybucji i biodostępności fluwastatyny.
Statins, including fluvastatin, which are HMG-CoA reductase inhibitors, are used as lipid-lowering drugs.It is assumed that statins can affect endogenous synthesis of polyunsaturated fatty acids, in which the key role is played by fatty acid elongases and desaturases. It has been found that fluvastatin reduces the mRNA level for Δ6D in rat’s hepatic tissue , but does not affect the level of mRNA for Elovl-5. Until now, no studies have been carried out on the influence of fluvastatin on the mRNA levels of key enzymes in PUFA synthesis in adipose tissue. The aim of this study was to evaluate the effect of fluvastatin on the level of mRNA for Elovl-5 and Δ6D in adipose tissue of rats. The adipose tissues of male Wistar rats, from experiments previously conducted at the Department of Biochemical Analytics UJ CM, have been used. The test group (n=6) was treated (for 7 days) with fluvastatin (at a dosage of 25 mg/kg b.wt). The control group (n=5) was given distilled water (solvent of a drug). Evaluation of mRNA for Elovl-5 and Δ6D in adipose tissue was performed using a semi-quantitative PCR method (reverse transcription-PCR).The mRNA level for Elovl-5 was higher while the mRNA level for D6D was lower than in the control group. However, the differences were not statistically significant. It can therefore be argued that fluvastatin at a dose of 25 mg / kg does not affect the level of mRNA for these enzymes in adipose tissue.The cause of tissue differences in the effect of fluvastatin on the mRNA level for Δ6D can be explained by the way of distribution and bioavailability of fluvastatin.
