Zastosowanie reakcji Gewalda do syntezy małocząsteczkowych inhibitorów białka USP2

Praca skupia się na zastosowaniu wieloskładnikowej reakcji Gewalda (G-3CR) jako jednej ze znanych metod syntezy pochodnych tiofenu do otrzymania inhibitorów białka USP2. Jest to reakcja organiczna obejmująca kondensację związku karbonylowego z α-cyjanoestrem, w obecności siarki elementarnej i zasady, czego efektem jest wytworzenie wielopodstawionego 2-aminotiofenu. Uzyskane w ten sposób podstawione 2-aminotiofeny należą do grupy bardzo istotnych struktur, które wykazują interesujące profile farmakologiczne, dlatego też są one celem wielu badań klinicznych. Wiadomo już, że otrzymane w wyniku reakcji Gewalda pochodne, posiadają zdolności inhibicyjne względem białka USP2. Specyficzna proteaza ubikwityny 2 (USP2) należy do rodziny deubikwitynaz, które rozcinają połączenia ubikwityna- białko przeznaczone do unieczynnienia. Dodatkowo USP2 współdziała z cykliną D1, proto-onkoproteiną, której nieprawidłowa nadekspresja jest związana z nowotworzeniem. Ze względu na swoją onkogenną funkcję, USP2 jest zatem ważnym celem w terapiach przeciwnowotworowych. Istotnym jest zatem znalezienie takiego inhibitora, który zablokuje białko USP2, zwiększy degradację onkoprotein i w ten sposób zahamuje rozwój nowotworu. W niniejszej pracy przedstawiono próby zmian właściwości inhibitora białka USP2, jakie podjęto celem uzyskania jego efektywniejszej formy. Jedną z metod było wprowadzenie do jego cząsteczki grupy zwiększającej rozpuszczalność związku, mianowicie morfoliny, natomiast drugą- zamiana pierścienia izoksazolu na pirazol.
The focus of this art is the use of the Gewald multicomponent reaction (G-3CR) as one of the well-known methods for the synthesis of thiophene derivatives to receive inhibitors of protein USP2. This is an organic reaction involving the condensation of carbonyl compound with α-cyanate ester in the presence of elemental sulfur and a base, resulting in the formation of multisubstituted 2-aminothiophene. Obtained by that way substituted 2-aminothiophenes belong to a crucial structures that have useful pharmacological profile and that makes them targets for a large number of clinical trials. It is already known that received via the Gewald reaction derivatives have the protein USP2-inhibiting capacity. Ubiquitin specific protease 2 (USP2) belongs to the family of deubiquitinating proteases which are responsible for the cleaving ubiquitin from proteins that should be inactivated. Additionally, USP2 interacts with cyclin D1, which aberrant overexpression is linked to oncogenesis. Because of its oncogenic function, USP2 is an important target for anti-cancer therapies. Therefore it is essential to find such an inhibitor which not only does block protein USP2, but it also increases degradation of oncoproteins, and in that way inhibits the spread of cancer. This thesis shows the attempt to change the properties of inhibitor of protein USP2 that have been taken in order to obtain the more efficient form of inhibitor. One of the methods was introduction of solubilizer compound ( morpholine) into molecule of inhibitor and the second one-the change of isoxazole on the pyrazole.
