Zastosowanie kobalaminy oraz kwasu foliowego jako nośników potencjalnych chemioterapeutyków na bazie rutenu

Leki oparte na kompleksach metali odgrywają znaczącą rolę w terapii antynowotworowej. Z uwagi na ograniczenia w stosowaniu cisplatyny i jej analogów, nadal poszukuje się nowych metalofarmaceutyków o większej selektywności działania. Jednym z ciekawszych połączeń są kompleksy rutenu(II/III), oferujące nowe perspektywy w leczeniu nowotworów. Poprawa selektywności i biodystrybucji stosowanych cytostatyków może zostać osiągnięta przez utworzenie kowalencyjnego adduktu nośnik – lek, który ulegałby rozszczepieniu po wejściu do komórki. Obecnie uważa się, że obiecującymi nośnikami substancji terapeutycznych mogą być witaminy (kobalamina i kwas foliowy) potrzebne do wzrostu komórek nowotworowych.Celem pracy było zbadanie możliwości utworzenia trwałych połączeń kompleksów rutenu(II/III) z potencjalnymi nośnikami transbłonowymi (cyjanokobalamina), o zwiększonym powinowactwie do komórek nowotworowych. W ramach pracy przeprowadzono syntezę adduktów nośnik – potencjalny lek i analizowano jej produkty oraz wyznaczono stałe trwałości otrzymanych kompleksów. Badania były prowadzone z wykorzystaniem elektronowej spektroskopii absorpcyjnej i emisyjnej oraz techniki HPLC. Dodatkowo metodami modelowania molekularnego określono parametry geometryczne i termodynamiczne produktów syntez i określono wpływ przyłączanego kompleksu rutenu na konformację pierścienia korynowego.
Drugs based on metal complexes play an important role in anticancer therapy. Due to the limits of use of cisplatin and it’s analogues, there is a need for developing new metalopharmaceuticals with improved selectivity of action. One of the most interesting connections that deserve special attention are complexes of ruthenium (II/III), offering new perspectives in the treatment of cancer. Improving the selectivity and biodistribution of chemotherapeutic drugs can be achieved by forming a covalent adduct carrier - drug which can be cleaved after entry into the cell. The aim of the study was to investigate the possibility of the formation of a stable binuclear complexes involving ruthenium (II/III) and a potential transmembrane carriers (cyanocobalamin). with increased affinity to the tumor cells. The synthesis of carrier – potential drug adducts was carried out. The obtained products were identified using both absorption and emission spectroscopy and HPLC technique. The kinetic and spectroscopic methods were applied in order to determine the stability constants. In addition, geometric and thermodynamic parameters of the expected products as well as the influence of the attached ruthenium complex on the conformation of corrin ring were estimated using molecular modeling methods.
