Płeć a aktywność fotosyntetyczna dwupiennego gatunku Rumex thyrsiflorus Fingerh. w warunkach in vitro i ex vitro

Rumex thyrsiflorus Fingerh. (szczaw rozpierzchły) jest gatunkiem dwupiennym, posiadającym heteromorficzne chromosomy płci: 14 chromosomów u roślin żeńskich (2n = 12A + XX) i 15 chromosomów u roślin męskich (2n = 12A + XY1Y2). W populacjach naturalnych R. thyrsiflorus obserwuje się zaburzone proporcje płci, z przewagą osobników żeńskich, co może być wyjaśnione przez płciowo-zależne różnice w tolerancji na stres. Celem podjętych badań, opartych na pomiarach aktywności fotosyntetycznej na podstawie fluorescencji chlorofilu a z fotoukładu II, było stwierdzenie, czy istnieją zależne od płci różnice w funkcjonowaniu aparatu fotosyntetycznego w warunkach kultur in vitro, podczas aklimatyzacji do warunków ex vitro (hodowle doniczkowe) oraz podczas aklimatyzacji do warunków polowych. Badane regeneranty uzyskano na pożywce MS+0,5 mg/l TDZ, drogą organogenezy pośredniej (indukcja pędów przybyszowych via kalus) z hipokotyli pobranych z 11- dniowych siewek. Proporcja płci (stosunek osobników męskich: żeńskich) wśród zregenerowanych roślin wyniosła 1: 5,8. Nie wykazano różnic statystycznie istotnych w wartości analizowanych parametrów fotosyntezy pomiędzy osobnikami męskimi i żeńskimi na poszczególnych etapach hodowli. Zanotowano natomiast różnice statystycznie istotne (p<0,0001) w aktywności fotosyntetycznej u osobników żeńskich pomiędzy analizowanymi etapami kultury. Różnice te dotyczyły wartości czterech parametrów: Fv/Fm, TFm, Area oraz PI total. Najbardziej stresogennym i wrażliwym dla roślin etapem eksperymentu, wpływającym na sprawność aparatu fotosyntetycznego, okazał się być transfer z hodowli doniczkowej (w warunkach laboratoryjnych) do warunków polowych. Wyniki eksperymentu uzyskane u Rumex thyrsiflorus, jako modelowej rośliny dwupiennej, mogą stanowić punkt wyjścia do podjęcia zaawansowanych fizjologiczno-biochemicznych badań dotyczących zależnych od płci reakcji na stresowe czynniki środowiska.
Rumex thyrsiflorus Fingerh. (thyrse sorrel) is a dioecious species with heteromorphic sex chromosomes: 14 chromosomes in female plants (2n = 12A + XX) and 15 chromosomes in male plants (2n = 12A + XY1Y2). In natural populations of R. thyrsiflorus, biased sex ratios, with females predominate are observed, which may be explained by sex-related differences in stress tolerance. The aim of this study was, predicated on the measurements of photosynthetic activity based on the fluorescence of chlorophyll a from photosystem II, to ascertain whether there are sex-related differences in the functioning of the photosynthetic apparatus under in vitro culture conditions, during acclimatization to ex vitro conditions (pot cultures) and during acclimatization to field conditions. The tested regenerants were obtained on the MS medium + 0.5 mg/l TDZ by indirect organogenesis (induction of adventitious shoots via callus) from hypocotyls excised from 11-day-old seedlings. The sex ratio (male:female ratio) among the regenerated plants was 1:5.8. There were no statistically significant differences in the values of the analyzed photosynthetic parameters in males and females at individual stages of experiment. On the other hand, statistically significant differences (p<0.0001) in photosynthetic activity in females between the analyzed stages of the culture were noted. These differences concerned the values of four parameters: Fv/Fm, TFm, Area and PI total. The most stressful and sensitive stage of the experiment, which affects the photosynthetic apparatus, was the transfer from pot culture (laboratory conditions) to field conditions. The results of the experiment obtained in Rumex thyrsiflorus, as a model of dioecious species, may be the starting point for advanced physiological and biochemical research on sex-dependent responses to environmental stress factors.
