Próby zwiększenia produkcji metabolitów wtórnych w kulturach pędowych Ruta graveolens L.

W ramach pracy badano wpływ stężeń regulatorów wzrostu i rozwoju roślin (auksyny - NAA i cytokininy - BAP) oraz dodatku prekursora (fenyloalaniny) na akumulację metabolitów wtórnych w pędowych kulturach wytrząsanych Ruta graveolens. Kultury prowadzono na dwóch wariantach podłoża wg Linsmaier i Skoog (LS) – LS 0,1/0,1 (0,1 mg/l NAA, 0,1 mg/l BAP) oraz LS 3/1 (3 mg/l NAA, 1 mg/l BAP). Do kultur prowadzonych przez okres 4, 5 i 6 tygodni dodawano fenyloalaninę w stężeniu 1,25 g/l (próby badane). Jednocześnie prowadzono próby kontrolne. Kultury likwidowano w 2, 3, 4 i 7 dniu od dodania prekursora.W ekstraktach z biomasy metodą HPLC oznaczono zawartość pięciu kumaryn: ksantotoksyny, bergaptenu, izopimpineliny, imperatoryny, skopoletyny. Z kolei w hydrolizatach wykazano obecność kwasów fenolowych: galusowego, protokatechowego, p-hydroksybenzoesowego, syryngowego, p-kumarowego, ferulowego oraz kumaryn: ksantotoksyny, bergaptenu, izopimpineliny, imperatoryny oraz marmezyny. Dodatkowo, w hydrolizatach stwierdzono obecność galusanu epigalokatechiny.Najwyższą całkowitą zawartość kwasów fenolowych (1,97 mg/g s. m.) stwierdzono w drugim dniu od dodania fenyloalaniny, po czwartym tygodniu prowadzenia kultur na podłożu LS 0,1/0,1 – w próbie badanej, natomiast maksymalna całkowita zawartość kumaryn (98,06 mg/g s.m.) oznaczono w drugim dniu od dodania fenyloalaniny, po piątym tygodniu prowadzenia kultur na podłożu LS 0,1/0,1 – w próbie kontrolnej. Najwyższą zawartość (0,28 mg/g s.m.) galusanu epigalokatechiny stwierdzono w trzecim dniu od dodania prekursora, po czwartym tygodniu prowadzenia kultur na podłożu LS 0,1/0,1 – w próbie kontrolnej. Dodatek prekursora wyraźnie zwiększa produkcję kwasów fenolowych (prawie 2-krotnie) i galusanu epigalokatechiny (około 16-krotnie). Nie stwierdzono istotnego wpływu dodatku fenyloalaniny na produkcję kumaryn.
The object of this study was to examine the influence of concentration of plant growth regulators (auxin – NAA and cytokinin - BAP) and addition of precursor (phenylalanine) on accumulation of the secondary metabolites in agitated, liquid, shoot in vitro cultures of Ruta graveolens. Cultures were supposed to grow on two different versions of medium, based on Linsmaier and Skoog medium – LS 0,1/0,1 (0,1 mg/l NAA and 0,1 mg/l BAP) and LS 3/1 (3 mg/l NAA and 1 mg/l BAP). Into the cultures, which was maintained for 4, 5 or 6 weeks, phenylalanine was added on a concentration of 1.25 g/l. To control probes sterile water was added. Biomass was collected 2, 3, 4 or 7 days after addition of precursor.The presence of coumarins: xanthotoxin, bergapten, imperatorin, isopimpinellin and scopoletin was identified by HPLC method in the methanolic extracts, whereas in hydrolysates phenolic acids such as gallic acid, protocatechuic acid, p-hydroxybenzoic acid, syringic acid, p-coumaric acid and ferulic acid were found. Additionally, the presence of coumarins: xanthotoxin, bergapten, imperatorin, isopimpinellin, marmesin and epigallocatechin gallate was indicated in hydrolysates as well.The highest total content of phenolic acids (1,97 mg/g d.m.) was obtained on the second day after addition of phenylalanine, after the fourth week of maintained on media LS 0,1/0,1 – in the test sample , whereas the highest total content of coumarins (98,06 mg/g d.m.) on the second day after addition, after the fifth week of maintained on media LS 0,1/0,1 – in the control sample. The highest total content of epigallocatechin gallate (0,28 mg/g d.m.) was obtained on the third day after addition of phenylalanine, after the fourth week of maintained on media LS 0,1/0,1 – in the test sample. The addition of precursor significantly increases the production of phenolic acids (almost 2-fold) and epigallocatechin gallate (16-fold). There was no significant effect of the addition of phenylalanine for the production of coumarins.
