Czy SLPI to CPP?

SLPI jest niewielkim białkiem kationowym występującym w wydzielinach nabłonków, między innymi tych wyścielających drogi oddechowe. Jest silnym inhibitorem proteaz, takich jak elastaza neutrofilowa, trypsyna i chymotrypsyna. Z tego względu przyjmuje się, że jego rolą jest ochrona tkanek nabłonkowych przed proteolizą w trakcie fizjologicznej odpowiedzi immunologicznej.Mój projekt dotyczył regulatorowej roli SLPI w układzie immunologicznym. Badania prowadzone w Zakładzie Immunologii wykazały, że SLPI hamuje wyrzut sieci neutrofilowych. Postawiliśmy hipotezę, że SLPI przekracza błonę komórkową, oraz potencjalnie również błonę jądrową neutrofili, i zatrzymuje formowanie się sieci na samym początku tego procesu. Pytanie, w jaki sposób możliwa jest jego internalizacja, pozostaje otwarte.Celem mojego projektu było znalezienie molekularnych podstaw dla internalizacji SLPI do neutrofili.W pierwszym etapie badań przeprowadziłam analizę bioinformatyczną sekwencji i opublikowanych struktur SLPI. Szukałam konserwatywnych reszt w sekwencji, analizowałam domeny, z których składa się SLPI, oraz własności białka. Ponieważ w bazie danych PDB nie ma dostępnej struktury przestrzennej domeny na końcu N SLPI, przeprowadziłam modelowanie homologiczne tej struktury, wykorzystując jako szablon domenę na końcu C SLPI.W drugim etapie, w oparciu o drożdżowy system ekspresji SLPI, opracowany w Zakładzie, przygotowaliśmy i oczyściliśmy fragmenty SLPI, pełniące potencjalnie różne funkcje. Wybór fragmentów był oparty o wyniki analiz bioinformatycznych.W trzecim etapie projektu otrzymane fragmenty SLPI zostały oznaczone biotyną, aby można było je wykorzystać do badań na neutrofilach z wykorzystaniem mikroskopii fluorescencyjnej. Celem było sprawdzenie, czy SLPI lokalizuje się w cytoplaźmie, czy też w jądrze neutrofili. Ponieważ biotynylacja białka zmienia jego własności, przeprowadziłam wstępny eksperyment porównujący internalizację SLPI biotynylowanego i niebiotynylowanego. Wyniki wskazują na to, że biotynylowany SLPI i jego fragmenty stanowią dobry model do dalszych badań funkcjonalnych.
SLPI is a small, cationic protein found in epithelial secretions. It is a potent inhibitor of human proteases such as neutrophil elastase, trypsin and chymotrypsin, and as such it is believed to protect epithelial tissues from proteolytic activity during the immune responses.My project concerned the regulatory role of SLPI in the immune system. The results obtained in the Department of Immunology showed that SLPI inhibits the formation of Neutrophil Extracellular Traps (NETs). We hypothesised that SLPI crosses the cell membrane, and possibly also the nuclei membrane of neutrophils, and inhibits the formation of NETs at its very beginnings. Therefore, the goal of my project was to find the molecular basis for the internalization of SLPI to neutrophils. Three steps were done towards the goal.Firstly, I did the bioinformatical analysis of the sequence and available structures of SLPI. I searched for the conserved residues in the sequence, analyzed the domain architecture and protein properties. Also, because there is no structure of the N-terminal domain of SLPI available in the PDB, I did the homology modeling of this structure, using the C-terminal domain as a template.Secondly, based on the SLPI expression system in yeast, that had been developed in the Department, we prepared the constructs of fragments of SLPI and purified them. The choice of constructs was guided by the results of the bioinformatical analysis.Thirdly, the purified constructs were tagged with biotin, to be used for the fluorescence microscopy assays in neutrophils. We planned to determine whether the signal from SLPI is concentrated in the cytoplasm, or co-localises with the signal from the cell nuclei. Because the biotinylation modifies the properties of the protein, I did the preliminary experiment to compare the internalization of biotinylated and unbiotinylated SLPI. The results showed that the biotinylated SLPI and its constructs are a valid model and may be used for future functional studies in neutrophils.
