Steroidogenna funkcja poowulacyjnych wzgórków jajonośnych mieszczących niezapłodnione oocyty.

W trakcie procesu folikulogenezy warstwa ziarnista pęcherzyka jajnikowego różnicuje się w subpopulację komórek muralnych, wyścielających ścianę pęcherzyka oraz otaczającą oocyt subpopulację komórek ziarnistych wzgórka jajonośnego. Związki wydzielane przez oocyt hamują proces różnicowania się komórek ziarnistych wzgórka w komórki steroidogenne. Ich przeznaczeniem jest stymulowanie rozwoju oocytu, a cechą charakterystyczną – obecność receptorów androgenowych. Dopiero w okresie okołoowulacyjnym komórki te zaczynają syntetyzować progesteron (P4). Przedstawiona praca miała na celu zbadanie wpływu testosteronu (T), antyandrogenu 2-hydroksyflutamidu (2-Hf) blokującego receptory androgenowe, testosteronu łącznie z hydroksyflutamidem (T+2-Hf), prolaktyny (PRL) oraz prolaktyny z dodatkiem testosteronu (PRL+T) na syntezę i uwalnianie P4 przez poowulacyjne wzgórki jajonośne (COC) szczura, mieszczące niezapłodnione oocyty. Celem badań była także immunolokalizacja w komórkach wzgórka jajonośnego enzymów steroidogennych: dehydrogenazy 3β-hydroksysteroidowej (3βHSD) biorącej udział w syntezie progesteronu, oraz cytochromu P450c17, koniecznego do syntezy androgenów. Przeprowadzono również immunolokalizację receptorów androgenowych (AR). Analiza zawartości steroidów w homogenatach poinkubacyjnych COC wykazała, że dominującym hormonem był P4, podczas gdy zawartość estradiolu (E2) i androgenów (A) była kilkakrotnie niższa. Jednakże inkubowane COC uwalniały do pożywki jedynie P4, zatrzymując E2 i A. Charakterystyczną cechą profilu P4 było to, że COC uwalniały większe ilości P4 do pożywki niż te zatrzymywane wewnątrz COC. Najwyższą sekrecję P4 obserwowano w inkubacjach z dodatkiem PRL + T. Nie stwierdzono stymulującego działania testosteronu na COC, natomiast obecność w pożywce 2-Hf lub 2-Hf + T obniżała zarówno sekrecję jak i zawartość P4, a rezultatem była najniższa sumaryczna wartość progesteronu. Przeprowadzona immunolokalizacja enzymów steoidogennych wykazała ekspresję 3βHSD, cytochromu P450c17 oraz receptorów androgenowych w poowulacyjnych wzgórkach jajonośnych, jednakże wyniki reakcji były podobne we wszystkich badanych grupach. Przeprowadzone badania dostarczyły nowych danych na temat nieznanej do tej pory zawartości steroidów w mikrośrodowisku poowulacyjnych wzgórków jajonośnych szczura.
During folliculogenesis the granulosa layer differentiates into the mural subpopulation, which lines follicular wall, and surrounding the oocyte subpopulation of cumulus granulosa cells. Secreted by the oocyte compounds inhibit differentiation of the cumulus granulosa cells into steroidogenic cells. The cells stimulate the oocyte growth and they characterize by the expression of androgen receptors. Only before ovulation the cells begin to produce progesterone (P4). The objective of these studies was to investigate the effect of testosterone (T), antiandrogen 2-hydroxyflutamide (2-Hf) blocking androgen receptors, both testosterone and 2-hydroxyflutamide (T + 2- Hf), prolactin (PRL) and both prolactin and testosterone (PRL + T) on the synthesis and release of progesterone by the rat postovulatory cumuli oophori (COCs) bearing unfertilized oocytes. The aim of the study was also immunolocalization in postincubated COCs steroidogenic enzymes: 3β-hydroxysteroid dehydrogense (3βHSD), responsible for synthesis of progesterone, and cytochrome P450c17, necessary for the synthesis of androgens. Also immunolocalization of androgen receptors (ARs) was performed. Analysis of steroid content of homogenized post-incubated COCs showed that in all investigated COCs P4 was a dominating steroid, while estradiol (E2) and androgen (A) contents were several times lower. However, incubated COCs released only P4, retaining E2 and A. A characteristic feature of progesterone profile of postovulatory COCs was that they released higher amounts of P4 into the incubation media than they retained this steroid inside respective COCs. The highest secretion rate and total amount of P4 were observed in incubations supplemented with PRL +T. Added testosterone did not stimulate P4 release or content but the presence of 2-Hf+T or 2-Hf alone decreased both progesterone secretion rate and content, resulting in the lowest total value of progesterone. Immunolocalization of stereoidogenic enzymes showed expression of 3β-HSD, cytochrome P450c17 and ARs in post-incubated cumuli oophori, however, results of immunoreactions were similar in all investigated groups. These results provided new data on unknown so far steroid contents in the rat postovulatory microenvironment of postovulatory oocytes.
