Analysis of activity of Gstk1 gene and its role in spermatogenesis

Niepłodność jest obecnie poważnym problemem, który z roku na rok dotyka coraz większej liczby ludzkości . Dane, opublikowane przez Światową Organizację Zdrowia (WHO) ukazują, że aż 10-12% par na świecie boryka się z problemem bezpłodności. Jednym z wiodących czynników powodujący problemy z płodnością są czynniki genetyczne. Naukowcy z Uniwersytetu Georga-Augusta w Getyndze, używając techniki nokautu genowego, stworzyli linie mysie z wielokrotnym nokautem. Nokautowi poddano od jednego do sześciu genów 1KO-6KO (knock-out). Nokaut pojedynczych genów nie dawał żadnego efektu, jednakże kumulatywny efekt nokautu sześciu genów ujawnił się pod postacią bezpłodności około 20% samców. Następnie porównano transkryptom jąder linii 5KO z transkryptomem jąder bezpłodnych myszy linii 6KO. W wyniku analizy zaobserwowano zmianę ekspresji 55 innych genów wytypowano je jako geny mogące być powiązane z płodnością u samców. W niniejszej pracy używając metody Western blot przeanalizowano białko GSTK1, które wykazywało obniżony poziom ekspresji u samców niepłodnych linii 6KO. Analiza wykazała brak istotnych statystycznie różnic w ilości produkowanego białka pomiędzy typem dzikim, płodnymi i niepłodnymi samcami linii 6KO.
Nowadays infertility is a serious problem which affects more and more people every year. Official data, published by the World Health Organization, shows that 10-12% of couples around the world are struggle with the problem of infertility. One of the leading factors in fertility problems are genetic factors. Scientists from the Georg-August University in Göttingen, using the gene knockout technique, using the gene knockout technique, created mouse lines with multiple knockouts. They knocked out from 1 to 6 genes 1KO-6KO (knock out). Single gene knockout had no effect, however the cumulative knockout effect of six genes revealed in the infertility in approximately 20% of males. Next, the testicular transcriptome of the 5KO line was compared with the testes of sterile 6KO mice. The result of these analyzes showed changes in the expression of 55 other genes, and they were selected as genes that could be related to male fertility. In this study, the GSTK1 protein, encoded by Gstk1 gene, which transcription showed a reduced level in infertile males from 6KO line, was analyzed using the Western blot method. The analysis showed no statistically significant differences in the amount of protein produced between the wild-type, fertile and infertile male of 6KO lines.
