Analiza ekspresji markerów mezenchymalnych komórek macierzystych CD29, CD44, CD90, CD105 w błonie śluzowej i mięśniowej macicy świni

Macica jest jednym z najbardziej plastycznych organów wszystkich ssaków łożyskowych. Ludzka macica w trakcie całego okresu w życiu kobiety, w którym jest ona zdolna do posiadania potomstwa, pod wpływem hormonów steroidowych ulega nieustającym, cyklicznym zmianom, a w trakcie ciąży powiększa swoje rozmiary nawet dziesięciokrotnie. Już w latach siedemdziesiątych ubiegłego wieku wysnuto hipotezę, jakoby za tak duże zdolności regeneracyjne tego organu odpowiadały komórki macierzyste, które po raz pierwszy wyizolowane i wstępnie zbadane zostały w 2004 roku.Celem niniejszej pracy było zbadanie lokalizacji w błonie śluzowej i mięśniowej macicy świni komórek wykazujących ekspresję markerów mezenchymalnych komórek macierzystych takich jak CD29, CD44, CD90 i CD105. Jako organizm modelowy wybrano świnię ze względu na podobieństwa morfologiczne i fizjologiczne macicy świńskiej i ludzkiej. Ponadto zbadano ekspresję innych markerów takich jak CD140b, CD45, CD31, Oct4 oraz antygenu jądrowego PCNA i hormonu greliny.W wyniku przeprowadzonych reakcji immunohistochemicznych na skrawkach mrożeniowych zobrazowano za pomocą mikroskopii fluorescencyjnej komórki wykazujące ekspresję antygenów CD29, CD44, CD90, CD105, CD140b, CD31, Oct4 i PCNA oraz greliny w określonych rejonach endometrium i miometrium macicy. Nie wykazano natomiast ekspresji antygenu CD45 będącego markerem komórek hematopoetycznych. W hodowli komórkowej in vitro komórki wykazywały ekspresję CD29, CD90, CD105, CD140b i Oct4.Lokalizacja komórek wykazujących ekspresję markerów mezenchymalnych komórek macierzystych w strefie takich struktur jak nabłonek powierzchniowy, gruczoły, rejon okołonaczyniowy, stroma endometrium i mięśniówka może sugerować udział tych komórek w zachowaniu homeostazy macicy świni.
The uterus is one of the most plastic organs in all placental mammals. Human uterus undergoes cyclic changes under the influence of steroid hormones during entire period of woman's life, in which she is capable of having children, and throughout the pregnancy when uterus increases its size even up to ten times. Already in the seventies of the last century scientists proposed hypothesis that the uterine stem cells are responsible for such a large regenerative capacity. However, these stem/progenitor cells were first isolated and investigated in 2004.The aim of this study was to determine the location of cells expressing markers of mesenchymal stem cells such as CD29, CD44, CD90 and CD105 in the endometrium and myometrium of the porcine uterus. Pig was chosen as a model organism due to physiological and morphological similarities of porcine and human uterus. Additionally, the expression of other markers such as CD140b, CD45, CD31, Oct4, nuclear antigen PCNA and ghrelin hormone was examined.As a result of the immunohistochemical reaction and imaging by fluorescence microscopy we identified cells expressing the antigens CD29, CD44, CD90, CD105, CD140b, CD31, Oct4 and PCNA and ghrelin hormone in specific regions of the endometrium and myometrium of porcine uterus. There was no CD45 (marker of hematopoietic cells) expression. In in vitro cell culture cells expressed CD29, CD90, CD105, CD140b and Oct4.Location of cells expressing mesenchymal stem cells markers in the vicinity of luminal and glandular epithelium, perivascular zone, endometrial stroma and myometrium may suggest their involvement in maintenance of porcine uterus.
