Badanie wpływu struktury chemicznej cząsteczek naturalnych i syntetycznych auksyn na właściwości modelowych błon biologicznych w kontekście ekotoksyczności regulatorów wzrostu roślin.

Przedmiotem pracy magisterskiej była analiza wpływu wybranych przedstawicieli hormonów roślinnych do których należą syntetyczne auksyny: kwas 2-naftoksyoctowy (BNOA) i kwas 2,4-dichlorofenoksyoctowy (2,4-D), oraz auksyna naturalna kwas indolilo-3-octowy (IAA) na modelowe błony lipidowe imitujące biomembrany komórek roślin i zwierząt. Celem prowadzonych badań było potwierdzenie ekotoksycznego wpływu tych regulatorów wzrostu roślin na badane modelowe układy lipidowe błon komórek ludzkich (erytrocytów i fibroblastów) oraz komórek roślinnych (liści pomidora oraz korzenia kukurydzy). Analiza wyników została oparta na charakterystyce izoterm ciśnienia powierzchniowego w funkcji średniej powierzchni na cząsteczkę zarejestrowane dla monowarstw sprężonych na subfazie zawierającej roztwory auksyn oraz ultraczystą wodę. Uzyskane wyniki wskazują, że roztwory auksyn o stężeniu 1*10-3 M silnie wpływają na charakterystykę badanych lipidowych monowarstw. Przeprowadzone badaniach wykazały silniejszy wpływ auksyny BNOA na badane monowarstwy, w porównaniu do auksyny naturalnej. Badania wykazały również, że wśród badanych modeli, błony komórek roślinnych są bardziej podatne na działanie auksyn w porównaniu z membranami komórek ludzkich. Uzyskane wyniki potwierdzają cytotoksyczne działanie auksyn przejawiające się głównie w obniżeniu kondensacji badanych monowarstw lipidowych.
The subject of the Master thesis was to analyze the influence of three representatives of plant hormones: synthetic 2-naphthoxyacetic acid (BNOA) and 2,4-dichlorophenoxyacetic acid (2,4-D) as well as natural indole-3-acetic acid (IAA) onto model lipid membranes mimicking both plant and animal cells. The aim of undertaken experiments was to elucidate the problem of ecotoxic influence of these plant growth regulators on studied systems mimicking lipid composition of the membrane of human (erythrocytes and fibroblasts) and plant cells (tomato leaf and maize roots). The analysis of the results was based on the characteristics of the surface pressure (π) – area (A) isotherms registered for monolayers spread on the subphase containing plant hormone and as a reference on the surface of pure water. The obtained results revealed that auxins applied in the solutions of 1*10-3 M concentration, strongly influence the characteristics of studied lipid monolayers. Studies showed that the synthetic plant hormone (BNOA) interacts with the investigated lipid monolayers stronger than the natural plant hormone (IAA). Studies proved that among the model biological membranes, the highest susceptibility toward auxin influence was found for the membranes of plant cells. The obtained results confirmed cytotoxic effect of auxins, revealed mainly in the decreasing of the lipid monolayers' condensation.
