Sprawdzenie korelacji między właściwościami elektrodonorowymi powierzchni implantu metalicznego a zdolnością do tworzenia biofilmu bakteryjnego

W bieżącej pracy wyznaczono zależność adhezji bakterii do podłoża tytanowego. Dzięki swoim właściwościom wszczepy wykonane z tytanu są bardzo często stosowane w implantologii. Wieloletnie badania i obserwacje tego materiału, zadecydowały o umiejscowieniu go w grupie tworzyw o strategicznym znaczeniu dla medycyny. Celem przewodnim pracy było sprawdzenie korelacji między właściwościami elektro donorowymi powierzchni implantu (praca wyjścia), a zdolnością do adhezji bakterii szczepu Staphylococcus aureus.Cel przewodni realizowano przez następujące cele szczegółowe:-preparatyka powierzchni materiałów stosowanych na implanty (płytki tytanowe); modyfikacja podłoży za pomocą procesów: wytrawiania i wygrzewania oraz elektro anodyzacji.-charakterystyka przekształconych próbek uwzględniająca właściwości fizyko-chemiczne, analiza składu oraz topografii.-zbadanie właściwości elektro donorowych - pomiary pracy wyjścia-analiza adhezji bakterii do podłoża oraz zdolności do tworzenia biofilmu przez wybrany szczep bakterii (gronkowiec złocisty – Staphylococcus aureus)
Self-assembled monolayers (SAMs) are molecular systems formed spontaneously by chemisorption to the selected substrate. SAMs are currently one of the most widely used systems in organic nanostructures. One of the most crucial feature of SAMs, which defines the usefulness of these structures in real applications, is stability of their chemical bonding to the substrate via the head group and the degree of their two-dimensional order. The type of head group has a significant effect on stability of a monolayer. In this master thesis two types of self-assembled monolayer were compared with either thiol or carboxylic acid head group deposited on silver surface. In the initial phase of this project, experiments were focused on optimizing monolayer synthesis by selecting a common solvent for both types of SAMs. Analyzes were made using infrared reflection absorption spectroscopy (IRRAS) and contact angle measurements (ACA). In the next step, using X-ray photoelectron spectroscopy (XPS), quantitative measurements were performed to confirm the purity of the obtained monolayers. Finally, to determine the impact of the head group on the stability of binding monolayers with the silver substrate, exchange experiments were conducted for both types of monolayers in the common solvent. The exchange process was monitored using IRRAS and ACA techniques. Based on this analysis, higher stability of SAMs with the carboxylic head group on silver substrate was concluded as compared to their thiol analogues.
