Celem niniejszej pracy była analiza mikrosporogenezy i mikrogametofitogenezy oraz ocena żywotności pyłku okazów Erigeron annuus (L.) rosnących na stanowisku na terenie kampusu 600-lecia odnowienia UJ.Obserwacje cytoembriologiczne wykazały zaburzenia w toku mikrosporogenezy. W metafazie I obserwowano pojedyncze chromosomy poza płytką, a także opóźniające się chromosomy w anafazie I. Najprawdopodobniej opóźniające się chromosomy prowadziły do powstawania mikrojąder. W wyniku zaburzeń mejozy po pierwszym podziale tworzyły się diady, triady oraz diady z mikrojądrem. Ponadto obserwacje wskazują, że na tym etapie część komórek kończy podział mejotyczny. Pozostałe podejmują drugi podział, który także był zaburzony. W metafazie II obserwowano pojedyncze chromosomy poza płytkami metafazowymi, a także opóźnione chromosomy podczas anafazy II.Zaburzenia mejozy, tworzenie monad, diad, triad, tetrad, pentad i heksad, prowadziły do powstania ziaren pyłku różnej wielkości.Żywotność pyłku wahała się od 60.9% do 19% w zależności od zastosowanego testu. Najwyższy procent żywotnego pyłku uzyskano w teście acetokarminowym , najniższy przy zastosowaniu fluorochromu FDA. W teście Aleksandra 30.3 % ziaren pyłku było zabarwionych na czerwono co wskazywało, że jedynie 1/3 pyłku jest żywotna. |

The aim of the present study was to investigate the course of microsporogenesis, and to analyze the pollen viability of Erigeron annuus (L.) growing in a natural habitat located on the Campus of the Jagiellonian University. Cytoembryological investigations showed disturbances in the course of microsporogenesis. At metaphase I, single chromosomes situated away from the equatorial plate were observed. Moreover, lagging chromosomes as well as chromosomes bridges were present at anaphase I. Probably the lagging chromosomes observed at telophase I were the reason for the formation of micronuclei. As a result of disturbances in meiosis, diads, triads and diads with micronuclei were formed after the first division. Moreover, observations show that some cells finish their meiotic division at this point. The others undergo second meiotic division, which is also disturbed. In metaphase II single chromosomes were observed away from the metaphase plate. There were also some lagging chromosomes visible in anaphase II.Disturbances in meiosis, the formation of monads, diads, triads, pentads and heksads led to the formation of grains varing in size. Pollen viability depended on the test applied. Acetocarmine test showed 60.9% of viable grains, Alexander test showed 30.3 %, the lowest amount of viable grains, 19%, was observed in FDA test. |
