Białko MCPIP1 nowy, negatywny regulator stanu zapalnego uczestniczy w ważnych procesach fizjologicznych, takich jak adipogeneza czy angiogeneza, a także przyczynia się do indukcji apoptozy. MCPIP1 reguluje ekspresję genów dzięki aktywności RNazy poprzez specyficzną degradację mRNA m.in. interleukiny 1β (IL 1β), interleukiny 6 (IL-6) swojego własnego, a także wielu pre-miRNA. Coraz więcej wiadomo o roli MCPIP1 w stanie zapalnym, lecz jego rola w nowotworzeniu pozostaje niewyjaśniona.W niniejszej pracy udowodniono możliwość uzyskania silnej nadekspresji MCPIP1 w linii komórkowej Caki-1 na drodze transfekcji plazmidowej. Następnie podjęto próbę stworzenia modelu stabilnej ekspresji MCPIP1 w linii komórkowej Caki-1 z wykorzystaniem wektorów retrowirusowych oraz z powodzeniem stworzono system indukowanej, stabilnej ekspresji z wykorzystaniem wektorów lentiwirusowych. Powstały model indukowanej ekspresji MCPIP1 może zostać wykorzystany do dalszych badań nad rolą MCPIP1 w procesie nowotworzenia i tworzenia przerzutów, jak również jego potencjalnym zastosowaniem terapeutycznym. |

MCPIP1 new negative regulator of inflammation is involved in important physiological processes like adipogenesis or angiogenesis and contributes to induction of apoptosis. MCPIP1 regulates gene expression via its RNase activity and specifically degrades mRNAs of among others interleukin 1 β (IL-1 β), interleukin 6 (IL-6), its own and many pre-miRNAs. Currently knowledge about MCPIP1 role in inflammation is rapidly increasing but its role in carcinogenesis remains unknown.Here I report feasibility of strong MCPIP1 overexpression in Caki-1 cell line using plasmid transfection method. Then an attempt was made to obtain a model of stable MCPIP1 overexpression in Caki-1 cell line using retroviral vectors and an induced stable overexpression system was successfully created using lentiviral vectors. New model of induced stable MCPIP1 overexpression may be used in further study of MCPIP1 role in carcinogenesis and potential therapeutic application of MCPIP1. |
