Study of spectroscopic and optical activity of selected xanthophylls in various environments

W pierwszej części pracy, wykonano badania struktury wybranych ksantofili: astaksantyny i kantaksantyny w różnych rozpuszczalnikach organicznych, przy użyciu spektroskopii Ramana. Otrzymane wyniki wykazały silną korelację pomiędzy liczbą falową pasm markerowych na widmach ramanowskich i polarnością rozpuszczalnika. Dodatkowo, zaobserwowano zmiany konfiguracyjne w badanych cząsteczkach, co związane było z częściową izomeryzacją trans-cis.W drugiej części pracy otrzymano supramolekularne agregaty enancjomerów astaksantyny (3R, 3'R) i (3S, 3'S), przez rozpuszczenie badanych związków w mieszaninie rozpuszczalników (aceton: woda) o stosunku 1:9, i 3:7. Dla uzyskanych agregatów wykonano szereg pomiarów spektroskopowych przy użyciu spektroskopii absorpcyjnej UV-VIS, ECD (elektronowy dichroizm kołowy), spektroskopii Ramana i ROA (ramanowska aktywność optyczna). Zarejestrowane widma porównano z widmami monomerów o takim samym stężeniu (10-5 M) w acetonie. Otrzymane widma pokazują, że dla obu izomerów optycznych astaksantyny w roztworze 3:7 aceton:woda, uzyskano J-agregaty, a dla (3S, 3'S) astaksantyny w roztworze 1:9 aceton:woda, powstały H-agregaty.Nie uzyskano żadnych sygnałów na widmach ROA monomerów, natomiast dla agregatów otrzymano pasma związane zarówno z chiralnością molekularną oraz supramolekularną. W środowisku wodnym cząsteczki astaksantyny organizują się tworząc trójwymiarową helisę z grupami hydroksylowymi skierowanymi na zewnątrz do środowiska polarnego (woda), natomiast niepolarny szkielet astaksantyny tworzy odseparowany od wody niepolarny trzon, stabilizujący strukturę agregatu.Jak dotychczas spektroskopia ROA uważana była za doskonałe narzędzie do określania konfiguracji absolutnej i konformacji małych chiralnych cząsteczek. Przeprowadzone badania pokazują jednak nowe możliwości użycia ROA. Po pierwsze, po raz pierwszy zarejestrowano widma RROA chiralnych supramolekularnych agregatów; po drugie, na widmach ROA uzyskano pasmo związane z transferem chiralności od asymetrycznego atomu węgla obok grup hydroksylowych do wiązania wodorowego z cząsteczką wody; i po trzecie, po raz pierwszy zarejestrowano i opicano pasma nadtonów na widmach ROA.
In the first part of the research, the structures of chosen xanthophylls: astaxanthin and canthaxanthin were studied in different organic solvents. Results showed a strong correlation between the wavenumber of the Raman marker bands, and the polarity of the solvent. Additionally, configurational changes of the studied molecules were observed including the partial trans to cis isomerization.In the second part of the study, (3R,3’R) and (3S,3’S) astaxanthin supramolecular aggregates, obtained by dissolving of the compounds in 1:9, and 3:7 acetone:water solutions, were studied by means of UV-VIS, ECD (Electronic Circular Dichroism), Raman and ROA (Raman Optical Activity) spectroscopies. Recorded spectra were compared with the spectra of the respective monomers in the same concentration (10-5 M) in acetone. Obtained spectra highlight that for both optical isomers of astaxanthin in 3:7 acetone:water mixtures, J-aggregates were successfully obtained, whereas for (3S,3’S) astaxanthin in 1:9 acetone:water, H-aggregates were formed.ROA spectra of monomers were not obtained, while the aggregates present vibrational modes due to both molecular chirality and supramolecular chirality. The main hypothesis is that the astaxanthin molecules organize forming a three dimensional helix with the hydroxyl groups facing the polar environment, whereas the apolar skeleton of astaxanthin is bent toward the inside, creating an apolar environment stabilizing for them. Until now ROA spectroscopy is considered a perfect tool for determining the absolute configuration and conformation of small chiral molecules. But this study has gone much more beyond this concept, in three different aspects: firstly, it is the first time RROA has been employed to investigate supramolecular chiral structures of enantiomers (astaxanthin aggregates), secondly, obtained ROA spectra display a band that is attributed to the transfer of chirality from the chiral carbon next to the hydroxyl groups to the hydrogen bond closest to him, and finally, it is probably the first time that overtones of ROA bands have been registered.
