Akumulacja metabolitów wtórnych w wytrząsanych kulturach pędowych Ruta graveolens ssp. divaricata (Tenore) Gams

Ruta graveolens ssp. divaricata (Tenore) Gams stanowi m.in. cenne źródło kwasów fenolowych oraz kumaryn.W ramach niniejszej pracy prowadzone były badania dotyczące wpływu stężenia roślinnych regulatorów wzrostu i rozwoju (NAA oraz BAP) na zdolność akumulacji kwasów fenolowych oraz kumaryn w kulturach wytrząsanych kulturach. Kultury te były prowadzone na czterech wariantach podłoża Linsmaiera - Skooga: wariant LS1 (1 mg/l NAA oraz 1 mg/l BAP), LS2 (2 mg/l NAA oraz 2 mg/l BAP), LS3 (3 mg/l NAA oraz 1 mg/l BAP) i wariant LS4 (0,1 mg/l NAA oraz 0,1 mg/l BAP). Związki zawarte w ekstraktach metanolowych oraz w hydrolizatach analizowane były metodą HPLC.W ekstraktach metanolowych stwierdzono obecność następujących kwasów fenolowych: kwasu galusowego, kwasu protokatechowego oraz kwasu 3,4-dihydroksyfenylooctowego.W hydrolizatach stwierdzono dodatkowo obecność kwasu p-hydroksybenzoesowego, kwasu syryngowego, kwasu wanilinowego oraz kumaryn: marmezyny i umbeliferonu. Najwyższą całkowitą zawartość kwasów fenolowych w ekstraktach metanolowych uzyskano na wariancie podłoża LS3 (2,49 mg/g s.m.), natomiast w przypadku hydrolizatów na wariancie LS4 (0,342 mg/g s.m.). Najwyższą zawartość kumaryn uzyskano na wariancie podłoża LS4 (1,28 mg/g s.m.). Badania dowodzą, że zawartość regulatorów wzrostu i rozwoju roślin w testowanych wariantach podłoża LS wpływa znacząco na skład ilościowy akumulowanych metabolitów wtórnych.
Ruta graveolens ssp. divaricata (Tenore) Gams is a valuable source of phenolic acids and coumarins.The object of this study was to examine the influence of concentration of plant growth regulators (NAA and BAP) on the accumulation of phenolic acids and coumarins in agitated cultures. Cultures were supposed to grow on 4 different versions of the Linsmaier - Skoog medium: version LS1 (1 mg/l NAA and 1 mg/l BAP), version LS2 (2 mg/l NAA and 2 mg/l BAP), version LS3 (3 mg/l NAA and 1 mg/l BAP) and version LS4 (0,1 mg/l NAA and 0,1 mg/l BAP). The compounds found in the methanolic extracts and hydrolisates were analised by means of the HPLC method.The presence of phenolic acids: gallic, protocatechuic and 3,4-dihydroxyphenylacetic acid was found in the methanolic extracts, whereas in hydrolisates phenolic acids such as: gallic, protocatechuic, 3,4-dihydroxyphenylacetic, p-hydroxybenzoic, vanillic and syringic acid were found. Additionally, the presence of coumarins: marmesin and umbelliferone was found in hydrolisates as well.The highest total content of phenolic acids in the methanolic extracts was obtained in the third version of the medium LS3 (2,49 mg/g d.m.), whereas in hydrolisates there was a version LS4 (0,342 mg/g d.m.). The highest total content of coumarins in hydrolisates was obtained in the fourth version of the medium LS4 (1,28 mg/g d.m.).Furthermore studies have shown that the concentration of plant growth regulators affects the quantitative composition of accumulated secondary metabolites.
