Aktywność krążeniowa nowych heteroaromatycznych pochodnych fenylopiperazyny

Celem mojej pracy była ocena aktywności przeciwarytmicznej oraz hipotensyjnej sześciu nowych, heteroaromatycznych pochodnych fenylopiperazyny.W tym celu określono powinowactwo badanych związków do receptorów adrenergicznych metodą radioreceptorową. Określono również ich wpływ na ciśnienie tętnicze krwi u normotensyjnych szczurów oraz na prawidłowy elektrokardiogram. W modelu arytmii adrenalinowej sprawdzono aktywność przeciwarytmiczną.Spośród przebadanych związków wszystkie wykazały powinowactwo do receptora α1-adrenergicznego. Cztery z nich tj. 107/1, 109/5, 107/4 i 68/2 wykazały aktywność przeciwarytmiczną w modelu arytmii adrenalinowej, z czego najskuteczniejszym okazał się związek 107/4. Żaden z przebadanych związków nie wpływał znamiennie statystycznie na elektrokardiogram, jedynie związek 109/1 zmniejszył częstotliwość pracy serca. Istotne działanie hipotensyjne wykazał tylko jeden z przebadanych związków tj. 107/1. Otrzymane wyniki badań mogą wynikać z właściwości α-adrenolitycznych analizowanych związków.Precyzyjne wyjaśnienie mechanizmu działania nowo zsyntetyzowanych pochodnych wymaga dalszych badań farmakologicznych.
The aim of my study was to evaluate the antiarrhythmic and hypotensive activity of six new heteroaromatic derivatives of phenylpiperazines.Using radioreceptor methods the affinity of tested compounds to adrenergic receptors was determined. For these compounds their effect on blood pressure in normotensive rats and normal electrocardiogram has been identified. In the model of adrenaline arrhythmia the prophylactic antiarrhythmic activity was examined.All of the tested compounds possess high affinity to α1-adrenergic receptor. Four of them: 107/1, 109/5, 107/4 and 68/2 showed antiarrhythmic activity in the model of adrenaline induced arrhythmia, the most effective compound was 107/4. None of the tested compounds had statistically significant effect on the electrocardiogram, only compound 109/1 significantly decreased heart rate frequency. One compound (107/1) showed significant hypotensive effect. The obtained results may be related to the α1-adrenolytic properties analyzed compounds.The precise explanation of the mechanism of action of newly synthesized derivatives requires further pharmacological studies.
