Synteza hybrydowych cząsteczek o potencjalnej aktywności GABA-ergicznej i powinowactwie do receptora 5-HT6

We wstępie niniejszej pracy magisterskiej przedstawiono budowę oraz funkcje receptora 5-HT6. W opisie uwzględniono przykładowe ligandy receptora 5-HT6, które obecnie cieszą się ogromnym zainteresowaniem ze względu na potencjalne zastosowanie w leczeniu lęku, depresji czy zaburzeń kognitywnych. W dalszej części pracy omówiono przekaźnictwo GABA-ergiczne, ze szczególnym uwzględnieniem roli, funkcji oraz metabolizmu głównego neuroprzekaźnika hamującego: kwasu γ-aminomasłowego. Dużą uwagę poświęcono aktywnym farmakologicznie estrowym pochodnym kwasu γ-aminomasłowego, w tym związkowi BL-1020. Znalazł się on w III fazie badań klinicznych i stał się wzorem w projektowaniu cząsteczek wpływających na aktywność GABA-ergiczną.Z uwagi na anatomiczne rozmieszczenie receptorów 5-HT6 na interneuronach GABA-ergicznych oraz badania farmakologiczne ujawniające synergistyczne działanie przeciwlękowe, zaobserwowane po równoczesnym podaniu selektywnego antagonisty receptora 5-HT6 i diazepamu, zaprojektowano serię związków łączących potencjalnie aktywność GABA-ergiczną z wpływem na receptory 5-HT6. W tym celu połączono fragment 4-(piperazyno-1-ylo)-1H-indolu, odpowiedzialny za interakcję z receptorem 5-HT6, wiązaniem estrowym z kwasem γ-aminomasłowym.W części doświadczalnej opisano szczegółowo pięcioetapową syntezę estrów GABA i alkoholowych N-pochodnych 4-(piperazyno-1-ylo)-1H-indolu. W wyniku przeprowadzonych prac otrzymano trzy chlorowodorki 4-aminobutanolanu N-pochodnych 2-[4-(1H-indol-4-ylo)piperazyn-1-ylo]etan-1-olu (1a, 1b, 1d). Z uwagi na szybką hydrolizę chlorowodorków 4-aminobutanolanu N-pochodnych 3-[4-(1H-indol-4-ylo)piperazyn-1-ylo]propan-1-olu (1e-h) nie udało się ich wyizolować z mieszanin reakcyjnych.Otrzymane struktury końcowe (1a, 1b, 1d) zostały następnie przebadane w kierunku powinowactwa do receptora 5-HT6. Największą aktywność wykazał związek 1d posiadający ugrupowanie benzenosulfonamidowe w pozycji N1 pierścienia indolu i etanolowy łańcuch połączony wiązaniem estrowym z kwasem γ-aminomasłowym.
In the introduction of this thesis the structure and functions of 5-HT6 receptor have been presented. The description includes examples of ligands 5-HT6 receptor, which currently are very popular because of potential use in the treatment of anxiety, depression and cognitive disorders. In the distal part of thesis the GABA-ergic neurotransmission, with particular emphasis on the role, function and metabolism of the major inhibitory neurotransmitter: γ-aminobutyric acid were described. A lot of attention was paid to the pharmacologically active ester derivative of γ-aminobutyric acid, including compound BL-1020. It has advanced to III phase of clinical trials and has become a model in designing of molecules that affect the activity of GABA-ergic.Due to the anatomical location of 5-HT6 receptors on GABA-ergic interneurons and pharmacological studies that reveal a synergistic antianxiety effect observed after concomitant administration of a selective antagonist of 5-HT6 receptor and diazepam, a series of compounds was designed linking potentially activity of the GABA-ergic transmission with the influence on receptors 5-HT6. For this purpose fragment 4-(piperazin-1-yl)-1H-indole, responsible for interaction with 5-HT6 receptor was combined, via ester bond with the γ-aminobutyric acid.In the experimental section the five-step synthesis of GABA esters and alcohol derivatives of N-4-(piperazin-1-yl)-1H-indole has been described in detail. As a result of carried research three hydrochlorides 4-N-aminobutanolanu of 2-[4-(1H-indol-4-yl)-piperazin-1-yl]ethan-1-ol (1a, 1b, 1d) were obtained. Due to the rapid hydrolysis of the 4-aminobutanolanu hydrochlorides of N-derivatives of 3-[4-(1H-indol-4-yl)-piperazin-1-yl] propan-1-ol (1e-h) could not be isolated from their reaction mixtures.The resulting final structures (1a, 1b, 1d) were then examined for the affinity for the 5-HT6 receptor. The highest activity was observed for compound 1d having a benzenesulfonamide at the N1 position of the indole ring and ethanol chain connected via ester group with γ-aminobutyric acid.
