Celem niniejszej pracy była analiza wpływu temperatury hodowli na skład barwnikowy aparatu fotosyntetycznego mezofilnej sinicy nitkowatejAnabaena PCC 7120 z rodziny Nostocales. Akseniczne hodowle prowadzono przez 14 dni, w warunkach fotoautotroficznych, na pożywce płynnej BG11-N, w temperaturach15°C, 23°C, 28°C i 37°C z ciągłym wytrząsaniem. Wzrost sinic hodowli monitorowano poprzez pomiar gęstości optycznej przy 700 nm oraz spektrofotometryczne oznaczanie poziomu białka, chlorofilu i karotenoidów. Analizę ilościową najważniejszych barwników aparatu fotosyntetycznego (chlorofil, miksoksantofil, echinenon, β-karoten) przeprowadzono metodą wysokosprawnej chromatografii cieczowej w odwróconym układzie faz na kolumnie chromatograficznej Agilent Eclipse Plus C18. Najszybszy wzrost Anabaena PCC7120 zaobserwowano w temperaturze 23°C. Stwierdzono istnienie pozytywnej korelacji pomiędzy szybkością wzrostu a akumulacją zarówno chlorofilu, jak i karotenoidów w aparacie fotosyntetycznym Anabaena. |

The aim of this work was the analysis of the temperature effect on the composition of photosynthetic pigments in a mesophilic cyanobacteria Anabaena PCC 7120 (Nostocales). Axenic cultures were grown for 14 days, under photoautotrophic conditions, in a liquid BG11-N medium, at different temperatures (15°C, 23°C, 28°C, 37°C) with a constant shaking. The growth of cyanobacteria was monitored by the measurements of the optical density at 700 nm as well as by spectrophotometric quantification of the total protein level and total carotenoid and chlorophyll contents. The detailed quantitative analysis of main photosynthetic pigments (chlorophyll, myxoxantophyll, echinenone, β-carotene) was performed by HPLC-based method, with Agilent Eclipse Plus C18 reversed-phase column. The fastest growth of Anabaena PCC7120 was observed at temperature of 23°C. The positive correlation between the growth rate and the accumulation of both chlorophyll and main carotenoids was demonstrated. |
